爪蟾发育早期尾部再生的机制研究

摘要动物的再生能力对组织器官的损伤恢复非常重要,但是,因为哺乳动物的再生能力有限,其中的机理并不十分清楚。本课题主要以非洲爪蟾为例探讨不同发育时期(34-47期)器官受损后再生修复的能力,结果表明,不同发育期下的尾部恢复有明显的差异。随着蝌蚪的发育,受损尾部的畸变率显著上升,尾部恢复的程度降低,所需的时间也相应延长,并导致存活率显著性下降。本课题选取34-35期的蝌蚪为例研究非洲爪蟾在发育早期尾部切除对其生存率、生长发育和尾部再生的影响及调节机制。将34-35期的蝌蚪经过尾部切除手术后暴露在组蛋白去乙酰化酶抑制剂中,其中TSA浓度为0、12.5、25和50nM;MS275浓度为0、2.5、5和10μM;MC1568浓度为0、2.5、5和10μM的培养液中。结果显示,抑制剂对蝌蚪尾部恢复起到抑制作用。通过分析,我们还得出了药物的最佳处理浓度为:TSA 12.5nM、MS275 2.5μM、MC1568 2.5μM。最后我们通过观察蝌蚪的行为学发现,随着组蛋白去乙酰化酶抑制剂处理浓度的上升,蝌蚪逃避光点的频率呈显著下降趋势。以上数据表明,组蛋白去乙酰化酶在蝌蚪尾部再生过程中起到非常重要的作用。71634

The regeneration ability is very important for the repair of the animal organs However, it is extremely limited for the mammalian regeneration ability and the underlying mechanism is not very clear. Here, we studied the ability of the regeneration and the repair of damaged organs in different developmental stages (stage 34-47) in Xenopus laevis. Our data showed that the recovery rate of the tail is significantly different  at developmental stages. With the development of the tadpoles, the abnormity rate of the damaged tail is significantly increased and the recovery of the tail is delayed, whereas the survival rate decreases significantly. We also studied the the regulation of tail regeneration at the early stages of development. The 34-35 tadpoles after tail resection with exposure to histone deacetylase inhibitors, such as TSA at 0, 12.5, 25 and 50nM; MS275 at 0, 2.5, 5 and 10μM; and MC1568 at 0, 2.5, 5 and 10μM medium. We find that the inhibitors blocked the recovery of the tail of the tadpoles. Furthermore, we obtained the ideal concentration of the drugs: TSA at 12.5nM、MS275 at 2.5μM, and MC1568 at 2.5μM. Finally, the avoidance behavior is significantly decreased with exposure to the histone deacetylase inhibitors in a dose-dependent manner. These data suggest that histone deacetylases may play a key role in the regeneration of the tail in Xenopus laevis.
