摘要如今,在农作物种植中,农药已投入广泛应用。而农药的应用对食品安全、环境污染等都提出了挑战。微生物修复对降解环境中的农药残余是一种方便自然的方法。该课题目标是找到能降解多菌灵这种农药的菌株,并对该菌进行鉴定和生长特性的研究。这个课题对环境保护有一定的意义。首先从新沂农药厂附近长期生产使用多菌灵的农用土壤取样,通过富集、驯化培养,分离得到一株多菌灵农药降解菌VU-G。该菌株形成的菌落呈圆形、黄色、光滑、没有凸起,边缘光滑。通过革兰氏染色,该菌株为革兰氏阳性菌。用分光光度计测定不同单因子变量下的OD值,得到该菌株生长最适盐浓度为7%,最适pH为9,最适通气量为150mL。通过观测该菌与八种营养物质在试管内反应的浑浊程度来测定该菌株的碳源,得出结论:该菌株能充分利用果糖,可以利用甘露醇、纤维二糖、乳糖和山梨醇,不能利用葡萄糖、蔗糖和维生素B1。通过上述特征和扩增测序,构建系统发育树,该菌株被鉴定为蜡样芽胞杆菌属的一个种。69926

Isolation, identification and growth characteristics of a carbendazim degrading strain UV-G

Abstract

Nowadays , pesticide has been widely used in the crop. And the application of pesticides has posed a challenge to food safety and environmental pollution. Microbial remediation is a natural method of the degradation of pesticide residues in the environment. The goal of this study is to find a bacteria can degrade carbendazim, identify it and do research on its growth characteristics, that it has some significance to environmental protection. Firstly, an efficient carbendazim degrading bacterial strain was isolated by continuous enrichment, screened from long carbendazim-treated soils and the sludge of wastewater in Xinyi insecticide factory. We named it VU-G. The colonies of strain VU-G are round, yellow, smooth, no bumps and with smooth edge. VU-G cells are gram-staining-positive. By determining the OD with spectrophotometer under different single factors, the optimum growing NaCl concentration is 7%, optimum growing pH is 9, optimum ventilation is 150mL. By observing the results of the reaction of bacteria and eight nutrients in vitro, we can determine the carbon sources of the bacteria. The conclusion is that the bacteria can make full use of fructose, make use of mannitol, sorbitol, cellobiose and lactose, unable to use glucose, sucrose, and vitamin B1. Afterwards, we amplificated, sequenced and constructed phylogenetic trees. Therefore, we identified strain VU-G is a species in Bacillus cereus.
