摘要:本课题是以藻蓝蛋白为原料,研究经高压处理后对于蛋白性质的影响。选定8MPa进行加压,分为4个加压时间段,以常压的藻蓝蛋白作为对照,首先对加压后的蛋白进行红外,荧光光谱扫描,分析其结构特性的改变,发现加压后的蛋白结构无明显差异,但荧光强度增强。然后对加压后的藻蓝蛋白进行抗氧化性、细胞增殖抑制性和研究环境条件对其影响的实验,用ABTS法测定藻蓝蛋白的抗氧化性,分析出随着其浓度的升高,抗氧化能力也不断增强;用MTT法研究出藻蓝蛋白的增殖抑制性随其浓度的升高而增强,且在加压时间为1.5h时抑制性最强。加压后的藻蓝蛋白起泡能力,乳化能力以及稳定性也较常压的藻蓝蛋白更好。在pH=7,温度为4℃,黑暗避光条件下储存的藻蓝蛋白稳定性最好。24123

The effect of high pressure on the property of phycocyanin
Abstract:The phycocyanin was selected as material, and the effect of high pressure on the phycocyanin was examined in this study. 8MPa and four treatment times were selected, the phycocyanin in ambient pressure was as control. First, the phycocyanin was scanned the spectrum of infrared ray and florescent after pressing to analyze the structure change. We fond the protein structures had no significant difference after pressing. However, the fluorescence intensity had increased. Then the pressurized phycocyanin was researched its antioxidant activity, antiproliferative activity and the influence of environmental conditions..The phycocyanin was used ABTS method for determining the antioxidant activity.. We analyzed that antioxidant activity capacity increased with the raised concentration. The phycocyanin was examined by MTT method. The results showed its antiproliferative was enhanced with the increase of the concentration, and the strongest inhibitory when pressurized time at 1.5 h. Foaming ability and emulsifying ability of high pressure was better than ambient pressure. The best stable environment conditions of phycocyanin are at pH=7,4℃ and under the dark.
