摘要在基因库中搜索到可以表达乙酰酶的基因tdac并且对其进行原核表达。通过气相色谱和高效液相色谱分析,我们证实TDAC酶可以将乙酰氨基葡萄糖去乙酰化成氨基葡萄糖和乙酸;用NaOH滴定乙酸含量测酶活力方法研究了TDAC酶学性质。研究表明:TDAC酶最适反应温度为80℃,最适反应PH为8.0, Co2+、Fe2+两种辅助因子对TDAC酶催化作用的促进作用最强。,TDAC酶在45℃水浴2h后,酶活性依然稳定,60℃水浴2h后酶活力明显下降。根据我们的文献调研,在国内外的氨基葡萄糖生产工艺的相关研究报道中,尚未见到酶法去乙酰化方面的研究。46735
According to literatures, we searched the tdac gene to express acetyl enzyme and expressed the tdac gene. We confirmed the TDAC enzyme can make acetylglucosamine acetylate glucosamine and acetic acid by gas chromatography and high performance liquid chromatography (HPLC) analysis, we studied enzymology properties of the TDAC enzyme with using tne method thant NaOH titrate acid content to measure enzyme activity of TDAC enzyme .This Study has shown that TDAC enzyme optimum reaction temperature is 80℃, the optimum reaction PH is 8.0, Co2+ and Fe2+ are two cofactors of TDAC enzyme thant have the strongest role to promote catalytic activity. In 45 ℃ water bath after 2 h, the TDAC enzyme catalytic activity remained stable, in 60 ℃ water bath after 2 h , the TDAC enzyme catalytic activity decreased obviously . According to our literature survey,we haven't seen a enzymatic acetylation research in those research reports of glucosamine production process from home and abroad.
