摘要目的:以NO供体巯基亚硝基-谷胱甘肽(S-nitrosoglutathione GSNO)作为工具药,以已构建的人鼻咽癌细胞(Human nasopharyngeal carcinoma cell, CNE-2)为模型,研究GSNO对CNE-2细胞增殖的影响,并加以形态学观察方法进一步论证。方法:采用GSH和NaNO2(1:1)在酸性避光条件下反应的方法合成GSNO,利用分光光度计法定量。采用MTT法检测GSNO对CNE-2细胞生长的影响;采用形态学方法(倒置显微镜、荧光显微镜)检查在GSNO作用下CNE-2细胞形态的变化。结果:MTT实验结果表明,同一作用时间内,GSNO各处理组対细胞生长的抑制率随药物剂量的增加而上升。同一浓度下,GSNO各处理组对细胞生长的抑制率均随作用时间的延长而增加。结论:GSNO能明显抑制人鼻咽癌的生长,且其抑制的效果与剂量和作用时间有关。75386
Objective: The NO donor nitroso glutathione thiol (GSNO) as a tool in medicine, human nasopharyngeal carcinoma cell line(CNE-2) has been constructed as a model, to study the effect of GSNO on the proliferation of CNE-2 cells, and to further demonstrate the method of morphological observation。 Method: GSNO was synthesized by the reaction of GSH and NaNO2 (1:1) under the condition of acid light avoidance。 The effect of GSNO on the growth of CNE-2 cells was detected by MTT method。 Morphological changes of CNE-2 cells were examined by morphological method (inverted microscope and fluorescence microscope)。 Results: MTT experimental results show, the same time, inhibit the growth of GSNO cells in each treatment group and control rate increased with the increase of the drug dose。 At the same concentration, the inhibition rate of GSNO in each treatment group increased with the prolonging of the acting time。 Conclusion: GSNO significantly inhibited the growth of human nasopharyngeal carcinoma, and its inhibitory effect was related to the dose and time of action。
