摘要: 制备自组装HA/RHB/DNA三元复合物非病毒基因载体,并对其进行表征及细胞学评价。通过CBA(N,N'-双(丙稀酰)胱胺),MBA(N,N'-亚甲基双丙烯酰胺)及AEPZ(1-(2-氨乙基)哌嗪)合成带正电荷的RHB,与负电荷质粒DNA进行自组装形成二元复合物RHB/DNA纳米粒,进一步通过静电包衣技术对所制备的纳米粒进行修饰得到HA/RHB/DNA三元复合物,对所制备的三元复合物进行表征及细胞学评价。 本文所制备的HA/RHB/DNA三元复合物最优比为5:1:5。在此条件下,细胞转染效率最高,毒性最低,并在体内可以稳定存在。 本文制备的HA/RHB/DNA三元复合物载体系统是一种安全,有效而且稳定的非病毒基因递送系统,为合理设计载体系统提供了新的方法,也为进一步进行体内研究奠定了基础。20137

Prepare, characterization, cellular evaluation of self-assembled HA/RHB/DNA ternary complexes non-viral systerm for gene delivery
Abstract : prepare self-assembled HA / RHB / DNA ternary complex non-viral gene vectors, and its characterization and cytological evaluation. CBA (N, N'-bis (acrylic acid) cystamine), MBA (N, N'-methylene-bis-acrylamide) and AEPZ (1 - (2 - aminoethyl) piperazine) with synthetic positive charge RHB, and negatively charged plasmid DNA self-assembly to form a binary complex RHB / DNA nanoparticles, the nanoparticles prepared further modified to obtain HA / RHB / DNA ternary complex by electrostatic coating technology, prepared ternary complex characterization and cytological evaluation. The results in this paper prepared HA / RHB / DNA ternary complex optimal ratio of 5:1:5. Under this condition, the highest transfection efficiency cells, low toxicity, and can be stable in vivo. prepared HA / RHB / DNA ternary complex vector system is a safe, effective and stable non-viral gene delivery systems for the rational design of a carrier system a new method, but also for further in vivo studies of the foundation.
