摘要: 嘧啶类化合物是一类含氮的杂环化合物,因其显示出良好的生物活性,在生物制药等方面有着广泛的应用前景。嘧啶类化合物在医药领域应用广泛,目前此类化合物作为中间体合成的药物种类繁多。有些药物应用广、疗效快、毒副作用小,且具有其它药物不可替代的作用,被广泛应用于临床治疗中。本文介绍了嘧啶类化合物的现状和应用,介绍了几种不同的合成嘧啶酮的方法。通过总结所查阅多方文献资料了后,确定了最终的合成路线。实验使用4-氯乙酰乙酸乙酯和5-巯基-1-苯基四氮唑作为初始原料,反应得到第一个中间产物。将这个中间产物与盐酸胍反应形成嘧啶酮结构。最后用4-氯甲基苯甲酰氯和嘧啶酮结构上的氨基反应,得到目标产物。此次实验的合成路线具有产率较高,后续处理方便的优点。50501

Synthesis of A Pyrimidine Pharmaceutical Intermediates
Abstract: Pyrimidine compounds are a class of hetero-cyclic compounds containing nitrogen, because it shows good biological activity in bio-pharmaceuticals have a wide range of applications. Pyrimidine compounds are widely used in the medical field, currently these compounds as intermediates in the synthesis of a wide variety of drugs. Some medicines wide application, fast efficacy and toxicity, it has an irreplaceable role of other drugs and has been widely used in clinical treatment. This paper describes the status and application of pyrimidines and several different methods of synthesis of pyrimidinone. Now after the adoption of the summary of the multi-party literature to determine the final synthetic route. Experiments using ethyl 4-chloroacetoacetate and 1-phenyl-5-mercapto-tetrazole as a starting material, we get the first intermediate product FETCL-6609. Then we use the FETCL-6609 and guanidine hydrochloride to get the second intermediate product FETCL-6610. At last we use 4-(chloromethyl)benzoyl chloride and FETCL-6610 to get the ultimate goal of product FETCL-6620. The experimental synthetic route have advantages of it has a high yield and convenient for subsequent processing.
