摘 要 随着现在人们的生活水平越来越高、所吃的食物与以前相比向肉类转变,还有一些药物的胡乱使用, 这些年来,原发性胆汁性肝硬化(primary biliary cirrhosis, PBC)的发病率每年都在上升,但是患病的人群的年龄却越来越小。在一些非常有前景的靶标以及与它们相关的药物正在被研究,目前研究处于临床状态的药物有大概30 个左右,其中以法尼醇X 受体(FXR)作为靶标的奥贝胆酸(Obeticholic Acid,OCA, 又名6-乙基鹅去氧胆酸)就是其中非常重要的一个。奥贝胆酸可以辅助或者替代熊去氧胆酸(ursodeoxycholic acid, UDCA)作为原发性胆汁性肝硬化患者的治疗药物,也能解决一部分非原发性胆汁性肝硬化患者对于熊去氧胆酸的不耐受性或者帮助治疗对于熊去氧胆酸无充分应答的患者。奥贝胆酸在2002年第一次被研发合成,近几年来,国内外学者对奥贝胆酸给予高度关注,开展了多方面的研究。就目前而言,奥贝胆酸具有很大的市场价值。本课题主要针对奥贝胆酸的工艺合成方面进行优化,旨在得到适合奥贝胆酸工业化生产和低成本的合成路线,以寻找更为合适的工艺合成方法,满足市场对其需求。89458
Abstract With the improvement of people's living standards, the changes in diet, and drug abuse, the incidence of primary biliary cirrhosis (PBC) has continually risen during the last ten years, and the mean age tends to be younger。 At present, primary biliary cirrhosis is recognized as the last disease, which has not yet been addressed。 Therefore, researchers are focusing to find biological target and novel drugs。 There are currently approximate 30 drugs in clinical research, and obeticholic acid (OCA, also known as 6-ethyl chenodeoxycholic acid) is one of potential new drug targeting the farnesol X receptor (FXR)。 Obeticholic acid can replace ursodeoxycholic acid (UDCA) as a therapeutic agent for patients with primary biliary cirrhosis (PBC)。 Obeticholic acid was first developed in 2002, and used in patients who have not adequately responded to or tolerated to ursodeoxycholic acid。 In recent years, domestic and foreign scholars have given a high degree of attention to obeticholic acid, and carried out a wide range of research。 It is significant that obeticholic acid shows a very great market value。 In this work, the production method of obeticholic acid was optimized aiming at its industrial production at a low cost to meet the market demand。
