加拿大一枝黄花组织总多酚的提取和抗氧化研究

摘 要探究加拿大一枝黄花叶片组织总多酚和多糖的含量与季节变化,确定该植物利用的最佳采集时期为7月,为进一步综合利用加拿大一枝黄花提供实验基础。采用乙醇浸提和超声波辅助法提取叶片中总多酚,以酒石酸亚铁法测定其物质含量,并采用不同方法测定组织中总多酚对DPPH、羟基自由基、过氧化氢等成分的清除作用,以考察该类物质的抗氧化活性。并采用三种不同型号的大孔吸附树脂以静态吸附与解吸附实验进一步纯化组织中总多酚。实验结果表明:加拿大一枝黄花不同月份的叶片中多酚含量在0。9-2 mg/g之间,以7月份叶片中的含量最高,多糖含量在11-208mg/g,以6月份组织中含量最高。总多酚浸提液表现出较好的抗氧化活性,不同浓度的多酚浸提液对DPPH自由基清除效果好于同浓度的Vc,而清除羟基自由和超氧阴离子的效果弱于同浓度Vc,两者清除过氧化氢能力相当。采用D101、AB-8、NKA-Ⅱ三种大孔吸附树脂对多酚物质的静态吸附与解吸实验表明,NKA-Ⅱ和D101型树脂的吸附率和解吸率分别达到47。58%和58。48%。静态吸附和解吸动力学研究结果显示,NKA-Ⅱ型树脂对多酚浸提液具有较好的吸附和解吸性能,静态吸附与解吸的最佳时间分别为3h和10min。87210 毕业论文关键词:加拿大一枝黄花;总多酚;抗氧化活性;大孔吸附树脂;静态吸附与解吸 Abstract The content and seasonal changes of total polyphenols and polysaccharose from the leaves of Solidago canadensis L。 were studied。 The polyphenols were extracted with ethanol by ultrasonic assisted method and its content was detected by ferrous tartrate method。 And the antioxidant activities of total polyphenols on scavenging different free radicals were also studied。 The results showed that the content of total polyphenols extracted from leaves of Solidago canadensis L。 varied from 0。9 to 2 mg/g, and the content of polysaccharose varied from 11 to 208 mg/g, which suggested that the optimum collecting period for utilization was July and June respectively。 The total polyphenol extract has a great performance of antioxidant activity。 The DPPH radical scavenging activity of different concentrations polyphenol of different months was better than that of the Vc at the same concentrations。 However its scavenging effect on hydroxyl radical and superoxide anion were inferior to Vc。 The hydrogen peroxide scavenging effect of the polyphenol extract was similar to Vc。 The result of static adsorption and desorption experiments on extracted polyphenol by 3 types of macroporous resins, D101, AB-8 and NKA-Ⅱ, revealed that the adsorption ratio of NKA-Ⅱ resins and desorption ratio of D101 resins reached to 47。58% and 58。48% respectively。 The static adsorption and desorption kinetics tests displayed that NKA-Ⅱ resin had a better adsorption and desorption performance on tissue polyphenol extract, the optimum time of adsorption and desorption were 3h and 10min respectively。 Keywords: Solidago canadensis L。 ; total polyphenols;antioxidative activity; macroporous resin; static adsorption and desorption 目 录 第一章 绪论 1 1。1研究背景 1 1。2植物多酚概述 1 1。2。1植物多酚的理化性质及生理功能 1 1。2。2植物多酚的研究 2 1。3加拿大一枝黄花研究概述 3 1。3。1加拿大一枝黄花的生物学特性 3 1。3。2加拿大一枝黄花的危害 4 1。3。3加拿大一枝黄花的防治与应用 (责任编辑:qin) |
