黄瓜(Cucumis sativus。 L)是一种重要的世界性经济作物,近年来已成为了植物分子生物学研究的一种好材料。我们前期的实验表明,黄瓜中的SPL13基因很可能参与了植株性别分化调控,但其具体功能尚未有明确的报道。
本研究利用模式植物拟南芥作为研究材料,考察黄瓜CsSPL13基因对拟南芥生长发育的影响:观测比较转基因与野生型植株幼苗的生长情况;测量成年植株抽薹、开花时间,开花时莲座叶数量;统计开花植株的高度、分蘖数和长角果数量等表型数据。我们获得了如下的实验结果:通过验证筛选异源转化的野生型拟南芥,获得10株35S::CsSPL13转基因纯合株系;黄瓜CsSPL13基因异源积累造成转基因拟南芥主根增长、抽薹时间提前、首朵花开花时间的提前。
我们的实验表明黄瓜CsSPL13基因影响了植株的发育和时相转变进程,可以促进植物发育,也可以促进开花。期望通过研究为进一步揭示其调控机制提供新的数据和参考,为探索更多它们在黄瓜发育过程中的功能机制提供了研究基础。
Abstract
The interaction of miRNAs and their targets is broadly involved in plant development, and in a fine-tuning regulation of gene expression。 The targets of miR156 are SPLs which belongs to a family of plant specific transcription factors (TFs)。 The interaction between them plays important roles in promoting plant development, phase change and flowering process。
Cucumber (Cucumis sativus。 L) is one of the popular worldwide economic crops, which is also becoming a good material in plant molecular biology research。 Our previous experiments have shown that the cucumber SPL13 gene may be involved in the regulation of plant sex differentiation, but the specific function is not yet clear。
In this study, we use the model plant Arabidopsis thaliana as research materials to examine the influence of cucumber CsSPL13 gene on the growth and development of transgenic Arabidopsis。 We compared the transgenic and wild-type plants, measured the adult plant bolting and flowering time, numbers of rosette leaves, plant height, number of tiller and siliques。 The main results are as follows: 10 homologous 35S::CsSPL13 transgenic plants were obtained; Ectopic expression of CsSPL13 in Arabidopsis led to elongation of primary roots and advanced bolting and flowering time。源Y于U优I尔O论P文W网wwW.yOueRw.com 原文+QQ75201-8766
Our study showed that CsSPL9 genes can influence the procedure of plant development and phase transition。 We believe this research provides basis for further study on molecular mechanism of miR156 and SPLs in cucumber。
