摘要斑马鱼是发育、遗传以及药物发现研究中常用的模式动物。作为重要的脊椎动物模式系统之一,斑马鱼的基因与人类基因具有高度同源性,中枢神经系统也与哺乳动物相似,故在研究化学药物对神经发育的影响时具有独特的优势。近年来由于社会高速发展,生产生活的各方面污染都越来越严重,苯系物在人类生存和周围环境中可被广泛检出,并对人体的血液、神经、生殖系统都具有较强危害。我们以DAT转GFP的斑马鱼为研究平台,利用震动切片、Confocal成像及行为学测定等技术对甲苯、二甲苯对斑马鱼幼鱼神经的毒性影响做初步研究。我们发现甲苯、二甲苯会对斑马鱼幼鱼神经的发育产生影响,特别是在数量和聚集度方面;另外,甲苯、二甲苯对斑马鱼幼鱼的行为学也有影响,经苯系物处理过后的斑马鱼幼鱼与对照组相比出现游动距离缩短的现象,行为能力减弱,行动迟缓。89028
Abstract Zebrafish is common model animal used in development, genetics and drug discovery。 As one of the important vertebrate model systems, zebrafish and human genes are highly homologous, central nervous system is similar to mammals, so it has an unique advantage of neural development in the research of chemical drugs。 In recent years due to the rapid development of social production, all aspects of life are more and more serious pollution, benzene can be widely detected in human survival and the surrounding environment, and do great harm to the body's blood, nervous and reproductive system。We take the zebrafish that DAT transferred GFP as a research platform,We discuss the preliminary study of Toxic effects of toluene and xylene on zebrafish larvae nerve, utilize techniques such as vibration slice, Confocal imaging and behavioral measurement。We propose that toluene and xylene affect the development of zebrafish larvae nerve, especially in terms of quantity and concentration。 In addition, toluene and xylene also affect the behavior of zebrafish larvae。 After the treatment of benzene homologues, the swimming distance of the zebrafish larvae is shorter than that of the control group, and the capacity for action is weaker。
