摘要当植物生长到一定年龄后,在适宜的外界条件下,营养枝的顶端分生组织(shoot apical meristem, SAM)就分化出生殖器官——花,最后结出果实。开花是植物从营养生殖转变为生殖生长的重要转折点。现有研究表明,中介体复合物(Mediator Complex)参与生殖生长等生命活动,在植物的发育调控中不可缺少。89625
为进一步探究MED10在植物生长发育中的功能,我们将其转入pCAMBIA1300-35S超表达载体中,构建pCAMBIA1300-35S-MED10超表达载体,利用农杆菌介导的转化法转基因得到阳性株系,以期得到纯合转基因株系,为研究该基因的功能奠定基础。
Abstract When plant grows to a certain stage, under appropriate external conditions, Vegetative shoot meristem (shoot apical meristem, SAM) differentiates into reproductive organs – flowers, and finally fruits。 Flowering is an important turning point in the transformation of plants from vegetative reproduction to reproductive growth,and flowering time is controlled by the intrinsic factors of the plant and environmental factors。 Research shows that in the model plant Arabidopsis, Mediator complex participates in reproductive growth and other life activities, so it is indispensable in the developmental regulation of plants。
In order to further explore the function of MED10 in plant growth and development, we transferred it into pCAMBIA1300-35S overexpression vector to construct pCAMBIA1300-35S-MED10 overexpressing vector。 Transgenic lines were obtained by Agrobacterium tumefaciens-mediated transformation to obtain homozygous transgenic lines, which laid the foundation for the study of the function of this gene。
