摘要:目的:通过测定蚂蟥冬眠不同时期细胞凋亡与自噬相关基因的表达,从基因组学的角度来研究冬眠蚂蟥细胞凋亡与自噬相关基因表达的变化 方法:通过荧光定量PCR技术检测蚂蟥冬眠不同时间AIF1、caspase-3、caspase-8、ATG3、ATG7、bcl-2、Beclin-1的表达的变化 结果:研究发现,在整个冬眠处理过程中,细胞凋亡相关的基因AIF1、caspase-3、caspase-8、bcl-2等整体表现为先降低后升高的变化趋势,在深冬眠时期的表达量达到最低值;细胞自噬相关的基因ATG3、ATG7、beclin1等整体表现为先升高后降低的变化趋势,在深冬眠时期表达量达到最高值。变化趋势明显,差别显著。结论:冬眠期间,蚂蟥细胞凋亡的水平降低,细胞自噬的水平升高。25769 毕业论文关键词:蚂蟥;冬眠;细胞自噬;细胞凋亡;基因表达
Study on Autophagy and Apoptosis regulation of Whitmania pigra in different time of Hibernation
Abstract:Objective: through the determination of expression in different periods of hibernation whitmania pigra apoptosis and autophagy related genes, changes from genomics perspective to study the expression of apoptosis and cell hibernation whitmania pigra autophagy related gene expression detection: through changes of the whitmania pigra hibernate fluorescence quantitative PCR technology, caspase-3 and caspase-8 in different time of AIF1, ATG3, ATG7, bcl-2, Beclin-1 the study found that: in the whole process of hibernation, apoptosis related gene AIF1, Caspase-3, Caspase-8, bcl-2 etc. overall showed a trend of decreased first and then increased, reaching the lowest value in the expression of deep hibernation period; autophagy related gene ATG3, ATG7 and Beclin1 for the overall performance trend increased and then decreased, reached the maximum expression in deep hibernation period. The trend of change is obvious, and the difference is remarkable. Conclusion: during hibernation, whitmania pigra apoptosis decreased, increased autophagy level.
