摘要:效应子是病原菌分泌到寄主细胞间或细胞内行使功能的一类蛋白分子,由病原菌基因组上相关的基因编码而成,研究人员在大豆疫霉(Phytophthora sojae)基因组中归类出了一个RxLR效应子家族,根据目前的研究结果,这个效应子家族是植物病原卵菌所特有的,该家族的基因在疫霉属(Phytophthora)的病原菌中大量存在,且许多RxLR效应子已被证明在疫霉菌的致病过程中发挥着重要的作用;但RxLR家族效应子之间基因序列高度分化,在不同的疫霉菌之间很难找到比较保守的RxLR基因。本课题使用生物信息学的方法对卵菌疫霉属clade7分支上的瓜类疫霉(P. melonis)RxLR效应子家族成员进行了预测,并将其与同为clade7分支上的大豆疫霉(P. sojae)RxLR效应子家族成员进行比较分析并找出种间存在的较保守的RxLR效应子。30818 毕业论文关键词:瓜类疫霉;大豆疫霉;RxLR效应子
The Analysis of RxLR Effector Family Between Two Phytophthora Pathogen Species’ Genome
Abstract:The effector is a class of protein molecules secreted by the pathogen to host cells, and is encoded by the genes associated with the genome of the pathogen. The researchers classified an RxLR effector family in the Phytophthora sojae genome and according to the current study, the RxLR effector family is unique to oomycete in plant pathogens, including bacterium and fungi. This family of genes has a large number of presence in pathogens of Phytophthora, it has been founded in many Phytophthora species and many RxLR effector have been proven to have an important role in the pathogenicity of Phytophthora. Although there are a large number of RxLR effector genes in pathogens of Phytophthora, the RxLR effector have highly differentiated gene sequence between different species, it is difficult to find a relatively conserved RxLR gene among different Phytophthora of oomycete. In this paper, we research the members of the RxLR effector family of P. melonis, which was predicted located in the clade7 branch of Phytophthora fimbriae, and compared the genes we researched with the P. sojae RxLR effector family members for comparative analysis through bioinformatics software and research methods. In this way, we could found some conservative genes between two Phytophthora species.

两种疫霉菌基因组中RxLR家族效应子基因的分析与比较
1 引言
大豆疫霉(Phytophthora sojae)是卵菌疫霉属(Phytophthora)的病原菌,引起大豆根茎腐病,会造成重大经济损失;瓜类疫霉(P. melonis)是由日本研究者桂琦一在1951年从黄瓜上分离鉴定得到的新种,在我国瓜类疫霉主要引起冬瓜和黄瓜病害。对这两个病原菌物种进行研究可以知晓与其生长、致病、增殖、越冬越夏相关的基因,并有针对性的在防控与治理手段上进行调整,从而有效减少生产经济损失。
效应子是病原菌分泌的可改变寄主植物细胞结构和代谢途径从而促进对寄主植物成功侵染或引发寄主防卫反应的一类外泌蛋白分子[1]。病原菌基因组中,有一类与植物编码的抗病基因相对应的受体基因,我们将这些基因称为“无毒基因”,其编码的蛋白称作“无毒蛋白”。RxLR效应子家族是在卵菌无毒蛋白的保守部分中鉴定出来的一类在N端第20-120位氨基酸区间中均具有”RxLR-dEER”基序[2],在C端氨基酸序列中部分具有长度约为21-30个氨基酸的W,Y保守域即WY-domain[3,7]的效应子,最早于2005年被发现[4],是植物病原卵菌所特有的。
