摘要:Immunoregulin HA是一种从拟黑腹牛虻(Hybomitra atriperoides)的唾液腺提取物中分离纯化出的生物活性肽,它由30个氨基酸残基组成,其肽段序列为GGVTGVTEFEPVDVSGEDYDSDEMDEDGRA,具有免疫抑制功能。之前的研究证明了Immunoregulin HA对免疫细胞细胞因子分泌的影响,为了进一步完善Immunoregulin HA对巨噬细胞的影响,我们对肽段的同源性进行分析,并以大鼠骨髓源巨噬细胞为材料,进行免疫抑制肽Immunoregulin HA对巨噬细胞精氨酸酶和产生NO的影响的研究。结果表明Immunoregulin HA可显著促进M2型巨噬细胞的精氨酸酶活,也可以极显著促进M1型巨噬细胞产生NO,即可促进iNOS的诱导合成,也可显著促进由LPS介导的M2型巨噬细胞炎症反应中的NO生成。巨噬细胞的精氨酸酶活和iNOS与其发挥抑制肿瘤和抗病毒的细胞毒作用有着密切关系。35242 毕业论文关键词:Immunoregulin HA;骨髓源巨噬细胞;精氨酸酶;iNOS
Primary research on how immunosuppressive peptide from salivary glands of the horsefly, Hybomitra atriperoides affect differentiation and function of mononuclear macrophage
Abstract: Immunoregulin HA is an immunopeptide which has been identified and characterized from salivary glands of the horsefly Hybomitra atriperoides. It is composed of 30 amino acid residues with the sequence of GGVTGVTEFEPVDVSGEDYDSDEMDEDGRA. Former research has proved its effects on cytokines secretions in LPS-induced inflammation. In order to investigate immunoregulin HA’s influence on macrophage, we study the homology of the peptide and do research on Arginase and NO generation of bone marrow derived macrophage(BMDM). The results show that immunoregulin HA can distinctly promote Arginase activity of M2 macrophage and can significantly distinctly promote NO generation of M1 macrophage. It means that immunoregulin HA can promote the synthesis of inducible nitric oxide synthase(iNOS). Meanwhile, immunoregulin HA can distinctly promote the NO generation of M2 macrophage under LPS-induced inflammation. Arginase and NO generation are closely related to the cytotoxic effect against viruses and tumor restraint of macrophage.
