摘要: 沃尔巴克氏体(Wolbachia pipientis)又称沃尔巴克氏菌,它广泛存在于节肢动物与线虫体内的细胞内共生菌,是迄今为止发现的在自然界分布最广的一类共生菌。本研究通过对沃尔巴克氏体wsp和ftsZ基因的PCR扩增来确定赤拟谷盗和杂拟谷盗体内的沃尔巴克氏体感染状况,并试图通过饲喂含有四环素的面粉对沃尔巴克氏体进行抑制消除,监测试虫的沃尔巴克氏体的抑制性消除状况,最后恢复普通面粉饲养试虫,观察其恢复状况。多次试验后发现,杂拟谷盗被沃尔巴克氏体感染,赤拟谷盗则没有被感染,在喂食四环素后,沃尔巴克氏体抑制性消失,但是用普通面粉饲养一段时间后沃尔巴克氏体又恢复了感染。本研究不但在理论上具有重要的研究意义,同时也为今后可能利用沃尔巴克氏体对拟谷盗进行预防和控制提供了理论依据。40898 毕业论文关键词:赤拟谷盗;杂拟谷盗;沃尔巴克氏体;感染状况;去除
Infection status and removal study of Wolbachia in Tribolium castaneum and Tribolium confusum
Abstract: Wolbachia is known as a symbiotic intracellular bacteria in arthropods and nematodes, and also it is the most widely distributed symbiotic bacteria in nature so far. In this study, we used PCR amplification of Wolbachia wsp and ftsZ specific gene to detect the Wolbachia in Tribolium castaneum and Tribolium confusum, then tred to remove the Wolbachia by feeding flour containing tetracycline and monitor removal conditions of tested insects, and finally restored the plain flour feeding and observed their recovery in tested insects. The results showed that Wolbachia existed in T. confusum, but not in T. castaneum. Wolbachia-negative beetles were getted by tetracycline flour treatment, but Wolbachia could resume growth and reproduction after feeding ordinary flour for a period of time. This study has both the important theory significance and the application value, which provides a basis for further research in T. castaneum and T. confusum.
