摘要:本文主要研究不同条件下柚子皮吸附材料对糖化酶、中性蛋白酶和木聚糖酶吸附性能的影响。采用实验中常用的单因素变量的实验方法,研究吸附材料对糖化酶、中性蛋白酶、木聚糖酶的适宜吸附时间、适宜吸附温度、适宜吸附浓度,并分别用Langmuir和Freundlich 两种吸附等温线模型对糖化酶、中性蛋白酶、木聚糖酶的吸附进行拟合。结果表明,柚子皮吸附材料对对糖化酶的吸附的最佳时间为15min,最佳吸附温度为323K,最佳吸附浓度为1.25mg/ml,对应的最佳吸附量为74.8 mg/g;对中性蛋白酶的吸附的最佳时间为20min,最佳吸附温度为313K,最佳吸附浓度为2.5mg/ml,对应的最佳吸附量为464.8 mg/g;对木聚糖酶酶的吸附的最佳时间为20min,最佳吸附温度为323K,最佳吸附浓度为2.5mg/ml,对应最佳的吸附量为396 mg/g。糖化酶、中性蛋白酶、木聚糖酶的△H、△S均为正值,说明吸附过程高温自发、低温不自发。以上研究结果表明柚子皮制备的材料有望成为糖化酶、中性蛋白酶和木聚糖酶吸附剂或固定化的载体。54480

Abstract:In this paper, the adsorption properties which the Citrus maxima bast (P1) adsorbent glucoamylase, neutral protease and xylanase were stutied. The effect of different factors, for example adsorption time, solution temperature, initial concentration of the enzyme were examined.The equilibrium data were fitted with Langmuir and Freundlich isotherm equations. The results showed that the best time of glucoamylase adsorpted is 15 minutes and the best adsorption temperature is 323K, the best adsorption concentration is 1.25mg/ml, corresponding to the optimum adsorption capacity of 74.8 mg /g. The best time of neutral protease adsorbed is 20 minutes and the best adsorption temperature is 313K, the best adsorption concentration is 2.5 mg/ml, corresponding to the optimum adsorption capacity of 464.8 mg /g. The best time of xylanase adsorbed is 20 minutes and the best adsorption temperature is 323K, the best adsorption concentration is 2.5 mg/ml, corresponding to the optimum adsorption capacity of 396 mg /g.To glucoamylase , neutral protease and xylanase, the positive values of the adsorption enthalpy △Hθ and the positive adsorption entropy △Sθ indicates that the adsorption being un-spontaneous in low temperature and being spontaneous in high temperature. The research results showed that P1 could be used as an ideal absorption material for the neutral protease and xylanase.
