摘要目前,我国肝癌的发病人数愈发增多,已经严重威胁到了人们的健康,临床上对肝癌的治疗方案还在不断完善中。本实验使用的木犀草苷是从药用植物秦艽中分离出来的,它是一种类黄酮天然化合物,实验目的在于研究木犀草苷对肝癌细胞增殖的影响。内容主要包括细胞增殖实验、活性氧实验和蛋白质免疫印迹实验,实验用到了CCK-8方法来确定木犀草苷对肝癌细胞增殖的影响。实验结果表明:木犀草苷能够明显降低细胞内活性氧的积累,使得活性氧水平降低,从而导致了NLRP3炎症小体表达减少,以及caspase-1失活,进一步抑制了IL-1β前体分子的成熟和释放。因此,木犀草苷能够对肝癌细胞的增殖产生抑制作用。通过我们对木犀草苷的研究,能够对其临床应用提供理论依据,为治疗肝癌提供新思路。69843

Effect of Luteoloside on Proliferation of Hepatocellular Carcinoma
Abstract
At present, the incidence of hepatocellular carcinoma in China is increasing. It has seriously threatened the health of people. The clinical treatment of liver cancer is still improving. This study aims to explore the luteoloside affection on human hepatoma cells proliferation. Luteoloside is a kind of flavone natural compounds. It’s isolated from the medicinal plant Gentiana. Study mainly includes cell proliferation assay, reactive oxygen species assay and western blot assay. CCK-8 method is adopted to determine the luteoloside’s effect on hepatocellular cells proliferation. Experimental results show that luteoloside can significantly reduce the accumulation of intracellular reactive oxygen species, so that the reactive oxygen species level reduces. As a result, NLRP3 inflammasome decreases in expression and caspase-1 inactivaties. This further inhibits the maturation and release of IL-1β precursor molecular. Therefore, luteoloside can suppress proliferation of hepatocellular carcinoma cells. Our study on luteoloside can provide the theoretical basis for its clinical application, it might benefit for new treatment of liver cancer.
