摘要微生物絮凝剂由于其高效,无毒且不形成二次污染的特点,被广泛应用于污水处理领域。【目的】 从河南花田土壤样品中分离筛选出具有絮凝活性的菌株,并对菌株进行分子鉴定以及产絮凝剂条件优化。【方法】 采用富集培养和平板分离法从土壤样品中分离菌株,根据菌株对高岭土悬液的絮凝活性筛选出絮凝剂生产菌。通过扩增并测序菌株16S rRNA对其进行分子鉴定。此外,从pH值、碳源、氮源等方面对絮凝剂产生条件进行优化。【结果】 从河南花田土壤样品中分离出一株具有絮凝活性的菌株,经鉴定,该菌株为弯曲芽孢杆菌。实验表明,该菌株在强碱性条件下仍可较好生长,乙酸钠和酸水解酪蛋白分别为碳氮源的条件下具有最佳生长率,乙醇和硝酸钾分别为碳氮源条件下具有最佳的絮凝活性。69840

Study In Polysaccharide Bioflocculant Producing Bacteria 

Abstract Due to the characteristics of high efficiency, nontoxicity and no secondary pollution, Microorganism flocculant is widely used in the field of wastewater treatment. [Objective] After isolating and screening of strain  which has flocculant activity from the flower fields’ soil samples in Henan, the molecular identification was performed and the conditions for flocculation are optimized. [Methods] Strains are isolated from soil samples in ways of the enrichment culture and the plate separation. Afterwards, floculant-producing strains are filtrated according to the strains that is for Kaolin suspension turbidity. The molecular identification is performed according to the 16S rRNA of strain. From pH, carbon source and nitrogen source, etc, the conditions of the flocculant are optimized besides. [Results] Strains are isolated from soil samples in ways of the enrichment culture and the plate separation. And this kind of strains is identified to be bacillus flexus. Moreover, some related experiments show that this kind of strains can grow better under the strong alkaline conditions. In the meantime, the sodium acetate and the acid hydrolyzed casein are endowed with the best growth rate as well as the ethyl alcohol and KNO3 possess the best flocculating activity under the conditions of the carbon sources and the nitrogen sources.
