摘要以二氯甲烷和吡啶分别为mPEG、PTS的溶剂,二者反应,将其产物溶解在DMF中,加入邻苯二甲酰亚胺钾继续反应。将第二次产物溶于无水乙醇,加水合肼,通N2,回流反应,从而得到mPEG-NH2。L-谷氨酸、苯甲醇和硫酸,70℃水浴反应,旋转蒸发,加碳酸氢钠溶液。将产物溶于THF中,加三光气,通N2反应,加正己烷沉淀。BLG-NCA溶解在氯仿中以mPEG-NH2为引发剂反应三天。将产物与二氯乙酸和25%的氢溴酸的醋酸溶液反应制得终产物,即具有两亲水性的聚L-谷氨酸-聚乙二醇单甲醚。通过红外光谱分析法对产物进行表征来分析确定其结构。63923

Title pH sensitive problem carrier material of synthesis of targeted drugs
Abstract With methylene chloride and pyridine as the solvent of mPEG and PTS respectively, mPEG and PTS is in chemical reactions. Firstly, dissolve the product in DMF and add phthalates formyl amine potassium . The solution is in the reaction again. Secondly ,the second product is dissoved in anhydrous ethanol and hydrazine hydrate is added ,adding nitrogen continuously in reflux reaction, thus we can get mPEG - NH2. L-glutamic acid, benzyl alcohol and sulfuric acid is mixed, heating the mixture in water bath at 70 ℃ . The product is taken a rotary evaporation again , and then add sodium bicarbonate solution into it . The product is dissolved in THF and three phosgene is added into it, adding N2 continuously. Last n-hexane is added. BLG - NCA dissolved in chloroform reacts for three days with mPEG - NH2 as the initiator . The product,dichloroacetic acid and 25% hydrobromic acid of acetic acid is mixed and react to get the end product -poly (L - glutamic acid polyethylene glycol single methyl ether, which has two hydrophilic property. The products were characterized with infrared spectrum analysis method to analysis and determine its structure.
