摘要: 本文4-羟基癸酸(盐)是由丙位癸内酯(γ-decalactone,GDL)进行逆向水解而制备。即以γ-癸内酯、水、氢氧化钠为原料,在100℃下利用皂化反应制备4-羟基癸酸钠,多次用氯化钙溶液进行复分解反应,经洗涤、抽滤、烘干得到纯度为99。4%的4-羟基癸酸钙,并用红外光谱仪、核磁共振色谱仪对产物的结构进行定性鉴定。传统认为生物转化制备GDL的代谢路径是蓖麻油酸通过4次β-氧化后形成4-羟基癸酸(4-HD),其作为内酯重要前体物经环化酶形成GDL。但4-羟基癸酸的化学性质不稳定在常温中性和偏酸的环境下容易环化成GDL,所以通过制备4-羟基癸酸钙加在含磷酸缓冲液(把钙盐缓慢转化成)培养基中,加入酵母粉10g/L进行48h生物转化生成GDL的方法,并用气相色谱仪测出产物的浓度。对其转化条件优化后得到起始PH为7。 6、底物浓度为2。37g/L、转化时间为60h时4-羟基癸酸钙生物转化效率最高。77003
