摘要:本文采用点滴法测定了fluralaner和氟虫腈对室内饲养的二化螟品系4龄幼虫的毒力,结果表明,在药剂处理24h和48h后,fluralaner和氟虫腈对二化螟的LD50值分别为0.022μg/头、0.025μg/头和0.003μg/头、0.004μg/头。在杀虫活性方面,处理48h后,两种药剂对二化螟4龄幼虫的毒杀作用都比24h明显增大。比较两者在48h时的LD50值可知,fluralaner对二化螟4龄幼虫的毒力大于氟虫腈,具有良好的杀虫效果。Fluralaner和氟虫腈的作用靶标均为二化螟GABA受体,故以二化螟的cDNA为模板,采用RT-PCR和RACE PCR技术得到了二化螟GABA受体的一个亚基的基因,并对其进行了相关的生物信息学分析,包括序列的一级、二级结构分析、进化树的构建与跨膜区预测,确定其为LCCH3亚基。LCCH3亚基的获得将为二化螟GABA受体的研究提供基础性信息。25566

The toxicity test of fluralaner on Chilo suppressalis and the cloning and analysis of fluralaner’s target receptor
Abstract: By topical application method, the toxicity activities of fluralaner and fipronil on Chilo suppressalis were tested in present study. The results showed that the LD50 values of fluralaner and fipronil were 0.022μg/head, 0.025μg/head and 0.003μg/head, 0.004μg/head after 24 hours and 48 hours of treatments, respectively. At the aspect of insecticidal activity, the toxicity of both insecticides on 4th instar larvae of C. suppressalis after 48 hours were higher than that of 24 hours. Compared the LD50 value, the toxicity of fluralaner was higher than fipronil’s on 4th instar larvae of C. suppressalis at 48h, which demonstrated that fluralaner had better insecticidal activity. As functional target of both fluralaner and fipronil, the GABA receptor of C. suppressalis was isolated as well. We use C. suppressalis’ cDNA as templates, a subunit gene of C. suppressalis’ GABA receptor was cloned by RT-PCR and RACE PCR. The primary and second structure analysis, the construction of the phylogenetic tree and the prediction of the trans-membrane region of the sequence were obtained by means of bio-information software and on-line tools. The cloned gene was confirmed as LCCH3 subunit gene at last. The obtaining of LCCH3 subunit would provide basic information for the study of C. suppressalis’ GABA receptor.
