摘要:本试验以‘神马’菊花CmBBX22基因为研究对象,利用构建好的pMDC43-CmBBX22 超表达载体,再以农杆菌介导的方法,侵染受体材料‘神马’的叶盘,对秋菊‘神马’进行转基因,经过抗性筛选后得到pMDC43-CmBBX22超表达的转基因菊花阳性株系,通过DNA水平PCR鉴定及qRT-PCR技术,对转基因苗进行检测,初步出鉴定出转基因阳性苗11个株系,对阳性苗扩繁炼苗成活扦插后,进行盐胁迫处理,发现CmBBX22转基因植株提高了耐盐能力。该研究为菊花的分子育种提供有用的候选基因,并为CmBBX22基因功能研究奠定了很好基础。25951 毕业论文关键词:CmBBX22基因;‘神马’菊花;转基因
Study on the function of CmBBX22 transgenic Chrysanthemum
Abstract:This research using chrysanthemum ‘Jinba’ as the material, the constructed over expression vector pMDC43-CmBBX22 was transformed into chrysanthemum mediated by Agrobacterium tumefaciens method with leaf disc. After transgenic over-expression positive plants pMDC43-CmBBX22 were screened, which was confirmed by RT-PCR. The positive seedlings were propagated, the cuttings were treated with salt stress, which showed a strong salt tolerant ability compared with the non-transgenic plants. Taking together, CmBBX22 gene plays an important role in salt tolerance of chrysanthemum.This study provided useful candidate genes for molecular breeding of chrysanthemum and laid a good foundation for the study of CmBBX22 gene function.
