摘要:铁蛋白(Ferritin,Ft)是一种广泛存在于生物体中的储铁蛋白,通常由24个亚基组成。壳-铁核结构、蛋白亚基间的大量盐桥和氢键,赋予了铁蛋白独特的理化性质。可逆组装、耐高温以及对pH敏感等特征,使其成为多功能的载体材料。通常会用来运载疏水/脂溶性的药物分子等,其中势必要引入有机溶剂。本文对马脾脏铁蛋白(HSF)进行除铁,对得到的去铁蛋白进行了一系列测试,并比较了去铁前后的紫外吸收和荧光发射光谱。研究了在有机溶剂的种类和量不同的情况下,去铁蛋白(APO)对5种染料分子:荧光素钠盐(FL)、罗丹明B(RhoB)、氯化尼罗兰(NB)、IR806、BODIPY类染料的吸附情况。

Title Study of the solvent resistance property of ferritin as a carrier material               

Abstract:Ferritins are iron storage proteins throughout a broad range of species. Generally, the typical ferritins are composed of 24 subunits. Considering the cage-like structure with an internal iron oxide core as well as sufficient salt bridges and hydrogen bonds between subunits, ferritins possess unique physical and chemical properties. The distinctive characteristics of ferritins, such as excellent heat resistance and pH dependent reversibly assembly, render them suitable candidates for multifunctional carriers. They are usually used to carry hydrophobic or hydrophilic drug molecules, which is bound to bring in organic solvents. In this paper, de-ironization of the as-received horse spleen ferritin is carried out in the first place. Then a clear colorless apo horse spleen ferritin(apoferritin) is obtained, followed by a series of property tests. In addition, UV spectra of the horse spleen ferritin before and after de-ironization is compared, so is luminescence spectra. The adsorption cases among apoferritin and five different dye molecules, including Fluorescein disodium salt(FL), Rhodamine B(Rho B), Nile Blue chloride(NB), IR806 and BODIPY, are studied in the presence of different types and amounts of organic solvents.
