摘要:豆科的模式植物——蒺藜苜蓿 (Medicago truncatula)目前是研究豆科植物和根瘤菌共生互作过程中信号调控的理想材料。植物中有一类特殊的小G蛋白ROP,其在囊泡的运输、信号的转导以及细胞的极性生长历程中起到了关键作用,而本文中的RIC家族是基于ROP的一种下游效应子。本研究通过建立高效的蒺藜苜蓿遗传转化体系,将RIC家族中的一个基因MtRIC8进行克隆及遗传转化,为进一步研究其在结瘤因子诱导的根毛变形过程中的功能以及研究豆科植物与根瘤菌共生互作信号调控机理奠定基础。实验结果综述为以下:(1)采用PCR技术扩增了RIC8基因,连接到pCAMBIA3301载体上构建了RIC8过表达载体;(2)通过侵染、共培养、诱导胚性愈伤及筛选、体胚发生等步骤获得了过表达植株;(3)对由第(2)步获得的植株进行PCR鉴定,结果表明遗传转化是成功的。26573 毕业论文关键词:蒺藜苜蓿、RIC8、转基因
Cloning and overexpression of MtRIC8 gene of Medicago truncatula
Abstract:As leguminous model plant,Medicago truncatula is an ideal material to explore signal control of symbiotic interactions mechanism between Leguminous plants and Rhizoid. ROPs are special small G proteins in plant, which significantly affect vesicle transport, signal transduction and polar growth of cell. RICs are the downstream effectors of ROP. This study established efficient Medicago truncatula genetic transformation system, cloning and genetic transformation MtRIC8 gene from the RIC family, laying a foundation for studying its function in the process of nodular factor induced-root hair deformation and signal regulation mechanism of the symbiotic interactions. The experimental results are following:(1) Using PCR amplified the RIC8 gene and linked it to the pCAMBIA3301 vector, building the expression vector; (2) Through infection, cocultivation, embryogenic callus induction, screening and somatic embryogenesis, we obtain the plant of gene overexpression; (3) Identified those plant by PCR, results show that the genetic transformation is successful.
