摘要:在油菜杂交种鉴定中需要鉴定杂交种的细胞质来源。鉴定细胞质可采用油菜线粒体基因组测序的方法,但是该方法过程复杂,成本过高,不易掌握。本研究尝试采用PCR鉴定方法,鉴定细胞质。通过提取油菜DNA,依据不同细胞质mtDNA的特异序列,设计引物,采用PCR方法扩增DNA片段,从而期望鉴定细胞质类型。叶片总DNA的PCR扩增结果表明,各细胞质类型样品中,均能检测出相同长度的片段,这说明用叶片DNA不能够鉴别差异的定细胞质。采用油菜线粒体DNA,通过PCR扩增实验,结果鉴别出了多态性条带,说明可区分细胞质类型。因此,细胞质鉴定需要采用线粒体DNA,用PCR方法鉴定,而不能够用叶片总DNA鉴定细胞质类型。34927 毕业论文关键词:油菜;细胞质;雄性不育
A Preliminary Study on Indentification of Cytoplasmic Male Sterility by PCR Method in Brassica napus
Abstract:In identification of Brassica hybrids, it is necessary to identify the cytoplasmic origin of Brassica napus parents. Determination of the mitochondrial genome of Brassica female parent is the best way to identify the cytoplasm origin, but it is complex, costs very much, and difficult to manipulate. This study used PCR method to identify the cytoplasm. By extracting the Brassica DNA, primers for the specific sequence of different cytoplasmic mtDNA were designed to amplify the DNA fragment by PCR method, and the cytoplasmic type may be identified by PCR products recogintion. Based on this principle, our experimental results showed that the cytoplasm origin may be identified by use of mitochondrial DNA. However, cytoplasm origin could not be detected by total DNA which includes the nuclear, chloroplastic and mitochondrial DNAs. This result might be explained by the mitochondrial evolution process. Therefore, we may conclude that cytoplasmic identification by PCR need to use the mitochondrial DNA.
