摘要:紫苏醛(Perillaldehyde)是提取于紫苏这种植物中的一种成分,被认为是紫苏挥发油抑菌的主要活性成分之一。本实验利用流式细胞仪通过对不同浓度的紫苏醛对黑曲霉细胞周期和细胞凋亡影响情况来研究紫苏醛的抑菌机制,主要由三个部分组成:紫苏醛对黑曲霉细胞膜甾醇含量影响,紫苏醛对黑曲霉细胞周期的影响,紫苏醛对黑曲霉细胞凋亡的影响。通过这三个部分的研究发现不同浓度的紫苏醛处理不同的时间对黑曲霉的作用不同。在0.5 µL/mL,0.75 µL/mL紫苏醛处理0 h,12 h,24 h,48 h时,黑曲霉阻滞于G0/G1期细胞数分别从26.8%升到33.6%,29.8%升到42.5%。而0.25 µL/mL,0.5 µL/mL紫苏醛处理分别3 h,6 h,9 h,也分别使黑曲霉的早期凋亡量分别从10.8%升至28.1%,17.5%升至41.1%。从本次实验我们也发现由于受到紫苏醛的影响,甾醇含量也受到影响,分别为未加紫苏醛处理的黑曲霉的甾醇含量约为0.50±0.13%,在0.125 µL/mL,0.25 µL/mL,0.5 µL/mL紫苏醛处理下的出黑曲霉细胞甾醇含量分别约为0.46±0.09%,0.32±0.03%,0.29±0.03%由此,可以得出紫苏醛对黑曲霉的作用是先从黑曲霉的细胞膜入手的,破坏黑曲霉的细胞膜,从而进入黑曲霉细胞内部,阻滞黑曲霉细胞周期和促进黑曲霉细胞凋亡等生理活动,阻止黑曲霉生长达到抗菌效果。通过本论文对紫苏醛抑菌机制的初步研究,能为以后更深一步地研究紫苏醛提供理论基础。同时,紫苏醛作为一种潜在的新天然防腐剂,拥有比合成的防腐剂更安全的特点,在防腐抗菌方面具有很大的应用前景。40834

Effect of Perillaldehyde on Aspergillus niger cell cycle and apoptosis
Abstract:Perillaldehyde is one of the component extracted from perilla.It is considered to be one of the main active ingredient from the perilla essential oil antibacterial. This experiment by flow cytometry is based on that different concentrations of perillaldehyde have effect on A. niger cell cycle and cell apoptosis situation to study the antibacterial mechanism of perillaldehyde,which is mainly composed of three parts: the effect of perillaldehyde on sterol content of cell membrane of A. niger, the effect of perillaldehyde on the cell cycle of A. niger,  the effect of perillaldehyde on the cells apoptosis of A. niger. Through the study of the three parts ,we found that perillaldehyde in different concentrations have different effects on A. niger under different time. The cell proportion of A. niger inhabited at G0/G1 phase rose from 26.8% to 33.6% ,29.8% to 42.5% under the treatment of 0.5 µL/mL,0.75 µL/mL perillaldehyde for 0 h,12 h,24 h or 48 h. The cell number of A. niger early apoptosis rose from 10.8% to 28.1%,17.5% to 41.1% under the treatment of 0.25 µL/mL,0.5 µL/mL perillaldehyde for 3 h,6 h or 9 h. From this experiment we also found that due to the influence of perillaldehyde, sterol content are also affected. The sterol content of cell membrane without perillaldehyde treatment of A. niger was about 0.50±0.13%, but under 0.125 µL/mL, 0.25 µL/mL, 0.5 µL/mL perillaldehyde treatment, the sterol content of A. niger were approximately 0.46±0.09%,0.32±0.03%,0.29±0.03%, so the function of perillaldehyde on A. niger is starting with the cell membrane of A. niger. It destructed the cell membrane of A. niger, then entered in the interior of the cell, in order to inhabit A. niger cell cycle and promote cell apoptosis, and inhibit the growth of A. niger and have the antibacterial effect. Through the preliminary study of perillaldehyde antibacterial mechanism, it got ready for deeper step research of perillaldehyde to provide certain theoretical basis. Perillaldehyde, as a natural preservative, is also healthier than synthetic preservatives, which has great potential development in terms of anticorrosive antibacterial.
