摘要:蛋白质是生物体中的大分子,是生物功能的直接体现者。新合成的蛋白质只有被运送到各自特定的亚细胞部位或分泌到胞外才能正确的行使其功能。研究蛋白质在细胞中的特异定位,可以增进对蛋白质以及与该蛋白互作的其他蛋白质功能的了解,帮助解释复杂的生命现象。蛋白质的亚细胞定位可以采取多种方法,如免疫荧光,基于荧光蛋白的亚细胞定位,免疫组织化学定位,免疫胶体金标记,免疫印迹等。本研究通过对免疫荧光方法中固定剂的选择和基于荧光蛋白的亚细胞定位中荧光蛋白基因位置的融和两种方法实验条件的探索,改进实验操作流程,以确定更适用于蛋白质亚细胞定位的方法。实验结果表明,免疫荧光法更适用于蛋白质亚细胞定位。39107 毕业论文关键词:亚细胞定位;免疫荧光;荧光蛋白(GFP)
The Comparison of Protein Subcellular Localization Methods
Abstract:Protein is a macromolecule in organisms, and it is the direct manifestation of biological functions. The proteins can function correctly when they are only transported to their particular subcellular site or secreted to the outside of the cell after they are synthesized. Study the specific location of the protein in cells can improve our understanding of the function of the protein as well as the other proteins which have interactions with it, and it is also conductive to explain the complex life phenomenon. There are many methods of protein subcellular localization,such as immunofluorescence, protein subcellular localization based on the fluorescent, immunohistochemical localization, immune colloidal gold labeling, western blot, etc. This study by immunofluorescence method in the choice of fixative and based on the fluorescent protein subcellular localization in fluorescent protein gene location of the harmony of two methods of experimental conditions to explore, improve the experiment operation process, to determine a more suitable for the method of protein subcellular localization. The experimental results show that the immunofluorescence technique is more suitable for protein subcellular localization.
