摘要本文尝试建立丙酮加热—超声提取分光光度法。选取超声提取分光光度法、热乙醇法、丙酮加热法、反复冻融法、热丙酮法、延时提取法6种常见的浮游植物叶绿素a的测定方法。本实验对杭州师范大学下沙校区教学楼D楼后的池塘水样进行叶绿素a含量的测定。实验分别从叶绿素a提取的浓度、测定数据的稳定性、实验操作的简便性等几个方面,比较了7种方法。实验比较了抽滤操作和离心操作对浮游植物叶绿素a测定结果的影响。实验对叶绿素a结果测定的单色法和三色法进行分析讨论。实验结果表明,抽滤操作和离心操作对测定结果无显著影响,鉴于抽滤操作耗时短,操作简便,可采用抽滤操作;三色法测量叶绿素a的浓度普遍高于单色法。从总体上讲,选取丙酮加热法测量叶绿素a的提取效率高、数据稳定性好,操作耗时短、简便。丙酮加热法可作为一种常用的浮游植物叶绿素a的测量方法。47205

Abstract
This study selected seven methods for the determination of chlorophyll a in Phytoplankton, including ultrasonic extraction spectrophotometric method hot ethanol, acetone heating, repeated freezing and thawing method, a thermal acetone, delay extraction and acetone heating - ultrasonic extraction spectrophotometry. Pond water sample was used to determine the content of chlorophyll a in Teaching Building D floor, Xiasha Campus, Hangzhou Normal University. The experiment compared effects of filtration and centrifugation operations on phytoplankton chlorophyll a measurement result, in accordance with extraction of chlorophyll a concentration, determination of the stability of the data and the simplicity of the experimental operation. The monochromatic method and the three color method for the determination of chlorophyll a were analyzed and discussed. Experimental results shown that, filtration and centrifugation operation on the determination results no significant influence, in view of the filtration operation time is short, simple operation, using vacuum filtration operation. Three color method for measuring the concentration of chlorophyll a is generally higher than the monochromatic method. On the whole, acetone heating method can be used as a common method for the measurement of phytoplankton chlorophyll a, because the method of measuring the chlorophyll a has high extraction efficiency, good data stability, and the operation is short and simple.
