摘 要:Prx V,也称为ALU的辅阻遏物1,过氧化物酶体的抗氧化酶,过氧化物酶,硫氧还蛋白还原酶,PRDX5和ACR1细胞质蛋白,是属Peroxiredoxin 2家族。本研究用生物信息学方法从氨基酸组成、理化性质、亚细胞定位、跨膜区和信号肽、二级结构、三级结构及功能域等方面对人Prx V蛋白进行了预测和分析。结果表明,Prx5蛋白分子量为22026.39,理论等电点(pI)为8.85,含有20种基本氨基酸,该蛋白质磷酸化以丝氨酸磷酸化为主,兼有苏氨酸和酪氨酸磷酸化且最可能为靶标蛋白,没有信号肽剪切点和跨膜区,α-螺旋占35.51%,扩展束占21.50%,β-转角占7.48%,无规则卷曲占35.51%。利用Geno3D同源建模方法对拟南芥A20/AN1锌指蛋白序列Q9SZ69.1进行模板匹配表明拟南芥A20/AN1锌指蛋白空间结构由3-4段不连续的α-螺旋片段和多段β-折叠组成。由分析可得此中蛋白质是一种线粒体靶标蛋白,通过与分泌蛋白的结合从而发挥作用。7417

Bioinformatic Analysis of Prx V Protein of
Structure and Function
Abstract:Study was carried out to predict and analysis of human Prx V protein with biological information from amino acid composition, method of physicochemical properties, subcellular localization, transmembrane region and signal peptide, secondary structure, tertiary structure and functional domains. Peroxiredoxin-5, also known as Alu corepressor 1,Peroxisomal antioxidant enzyme, Thioredoxin peroxidase PMP20, Thioredoxin reductase,PRDX5 and ACR1, is cytoplasm protein which belongs to the peroxiredoxin 2 family. Prx5 protein with molecular weight of 22026.39, theoretical isoelectric point (pI) of 8.85, contains 20 kinds of essential amino acids, the protein phosphorylation at serine phosphorylation, both threonine and tyrosine phosphorylation and most likely to target proteins, no signal peptide and transmembrane region, helix accounted for 35.51%, expanded beam 21.50%, β - turn accounted for 7.48%, accounting for 35.51% of random coil.The Arabidopsis A20/AN1 zinc finger protein sequence Q9SZ69.1 template matching show that Arabidopsis A20/AN1 zinc finger protein spatial structure by 3-4 discontinuous alpha helical segments and multi beta folding by Geno3D homology modeling method.By the analysis of protein in the mitochondria is a target protein, combined with secretory protein can play a role.
