摘要此次实验的宗旨是研究验证ACTA1(α-肌动蛋白1)基因在徐淮山羊(胎羊)不同组织(心、肝、脾、肺、肾、肠、背肌、腿肌)中的表达差异,利用的手段是使用实时荧光定量PCR的技术。这为了以后研究ACTA1基因在有关胎羊的徐淮山羊上的问题打下基础。实验操作的步骤是:第一,采取徐淮山羊胎羊的心、肝、脾、肺、肾、肠、背肌、腿肌等部位的组织若干份;第二,提取八个组织总的RNA,并测量与记录RNA的浓度;第三,利用ACTA1基因特异性引物将各个部分组织的RNA逆转录成cDNA;最后使用PCR技术来检测其表达谱。实验最终结果表明:ACTA1基因在八个组织内均有表达,但存在表达差异。其表达量由高到低依次为背肌(13.177070)>腿肌(6.431454)>心(1.00000)>肝(0.466959)>肾(0.381187>肺(0.118584)>脾(0.086988)>肠(0.071708)。用心肌作为参照,ACTA1基因在背肌和腿肌中的表达量是极显著的,在脾、肺和肠中的表达量是显著,在肝和肾中表达不显著。此结果可以作为今后对徐淮山羊(胎羊)ACTA1基因研究的实验基础和理论依据。51005

The study on different expression of ACTA1 gene in fetal lamb organs of Xuhuai goat
Abstract

The experimental results show that ACTA1 genes are expressed in eight in the organization are, but to express differences. The expression quantity order from high to low is back muscle (13.177070) > leg muscle (6.431454) > heart muscle (1.000000)> liver (0.466959)> kidney (0.381187) > lung (0.118584) > spleen (0.086988) > intestine (0.071708). With myocardial as a reference, ACTA1 amount of gene expression in back and leg muscle is very significant, the amount of expression in the spleen, lungs and intestine is significant, no significant expression in the liver and kidney. Experimental results can be used as the future of Xuhuai goats (fetal lamb) ACTA1 gene research experimental foundation and theoretical basis.
