摘 要:以水稻种子为材料,采取MS培养基,研究了乙醇处置水稻种子的时候对水稻种子抽芽的影响、次氯酸钠浓度对水稻种子萌生的影响和它们的灭菌结果,发明乙醇处置时候越长,出芽率越低,出芽时候越长且灭菌结果较次氯酸钠差;次氯酸钠浓的灭菌结果最佳且对种子的影响较小。还研究了多效唑浓度、蔗糖浓度、琼脂浓度、pH值对水稻苗的影响。结果表明:跟着多效唑浓度的增高,表现出显著的矮化结果;蔗糖浓度过高,会使水稻根系发生延缓;琼脂浓度过高影响水稻扎根,从而影响水稻的正常生长;当pH值低于5时会影响培养基凝固,还会影响水稻生根,当pH值为7时,更有利于水稻根的生长。因此,最适水稻种子生长的培养基为:基本培养基+5-15mg/L多效唑+30g/L蔗糖+7g/L琼脂,最适pH值为7左右。67936

Abstract:Rice seeds as materials, using MS medium were studied ethanol time rice seed on rice seed germination, sodium hypochlorite concentration on rice seed germination and their sterilizing effect, found that the longer the ethanol processing time, budding rate the lower the longer the budding time was better than sodium hypochlorite and poor sterilization; sterilization concentrated sodium hypochlorite best and less impact on the seeds. MET also studied concentration, concentration of sucrose, agar concentration, pH value of rice seedlings. The results showed that: with the increase of paclobutrazol concentration showed significant dwarfing effect; sucrose concentration is too high, the occurrence of rice root delay; agar excessive concentrations of rice roots, thus affecting the normal growth of rice; when the pH is below 5:00 will affect the medium solidified, will also affect rice root, when the pH value is 7:00, more conducive to the growth of rice roots. Therefore, the optimum medium for the growth of rice seed: basic medium + 5-15mg / L MET + 30g / L sucrose + 7g / L agar, the optimum pH value of about 7.

1 前言
水稻作为我国重要的的粮食作物,有着悠久的栽培历史。而且,水稻有着非常丰富和深入的基因组研究的基础,是一种模式作物[1],可以提供各种各样的研究。水稻作为粮食作物,除可用做食品,还可在工业和医学方面用到,可见水稻对我们生活有着重要影响[2]。因此为了方便进行对水稻的研究,培育水稻试管苗就显得很重要。为了使水稻得到更好的生长,种子的灭菌和培养基的配制就显得非常重要。本次实验采用的是MS培养基,通过改变培养基中的pH值、蔗糖和琼脂浓度以及多效唑的含量,观察并对比水稻种子在不同培养基中生长情况。
