摘要采用常规细菌分离和纯化的方法,从浙江附近海域海底表层沉积物中分离出两株中等程度的嗜酸细菌,分别命名为A和B。借助大肠杆菌和革兰氏染色对两株菌的大小进行了估测,对两株细菌的16S rDNA进行测序,建立系统发育进化树,初步判断其属种和亲缘关系。在随后的生理生化的实验中,研究两株菌在不同温度和不同pH下的生长状况。 A菌是革兰氏阳性菌,呈不规则杆状,大小为(0.5-1.5)μm×(2-5.5)μm ,最适生长温度是25 ,最适生长pH是值5.3,A菌和Nocardioides simplex(AF005009)的16S rDNA相似度最高,其同源性98.29%。B菌株是革兰氏阳性菌,近似卵圆形或椭圆形,大小约1.5-3.5μm,最适生长温度是30 ,最适生长pH值是5.2,B菌和 Marinococcus tarijensis(JQ413413)的16S rDNA高度相似,同源性达97.74%。68848

Isolation and identification of two strains of acidophilus bacteria from Zhejiang sea area
Abstract
Two strains of moderately acidophilic bacteria were isolated from the submarine surface sediments of the sea area near Zhejiang, which were isolated and purified by conventional methods, and were named as A and B, respectively. With the help of E. coli and gram staining to the estimate of the size of the two strains of bacteria, two bacterial 16S rDNA wear sequenced and the phylogenetic tree of the preliminary wear established to judge their species and genetic relationship,respectively. In the subsequent physiological and biochemical experiments, the growth conditions of two strains of bacteria at different temperatures and different pH was studied. A bacterium is a gram-positive bacteria, irregular rod-shaped, size (0.5-1.5) μm × (2-5.5) μm, the optimum growth temperature is 25 , the optimum growth pH value is 5.3, A and bacteria Nocardioides simplex (AF005009) have highest similarity in 16S rDNA ,their homology are 98.29%. B strain is gram-positive bacteria, approximately oval or elliptical shape, about the size 1.5-3.5μm, optimum growth temperature is 30 , the optimum growth pH value is 5.2, B bacteria and Marinococcus tarijensis (JQ413413) has high degree of similarity in 16S rDNA, their homology are 97.74%.
