摘要:本课题选取猴头菇子实体和香菇子实体,通过热水浸提、浓缩、分离、纯化、透析、冷冻干燥实验制得样品。用苯酚硫酸法测得猴头菇多糖和香菇多糖含量分别为75.2%(±0.64%)、 80.0%(±0.63%),通过高效液相色谱分析得到样品中的多糖较纯,用红外光谱分析出样品中多糖的结构为β-吡喃多糖。利用β-葡聚糖试剂盒方法测量样品中猴头菇多糖和香菇多糖中β-葡聚糖的含量分别为64.94%(±1.60%)和70.62%(±0.48%),利用离子色谱法测得这种工艺提取的猴头菇多糖样品的单糖组成为半乳糖、葡萄糖、木糖和甘露糖,香菇多糖样品的单糖组成为果糖、木糖、甘露糖、半乳糖和葡萄糖。利用BCATM试剂盒法测得猴头菇多糖样品和香菇多糖样品中蛋白质含量分别为4.84%(±0.12%)和8.68%(±0.20%)。将猴头菇多糖和香菇多糖分别与RAW 264.7巨噬细胞共同培养一段时间后,观察得出这两种多糖均具有一定的抗肿瘤效果。42890

preparation and primary structure analysis of Hericium / letinous edodes beta-glucan
Abstract :This topic selected fruitbodies of Hericium erinaceus and Lentinus edodes ,and obtained the samples by hot water extraction, concentration, separation, purification, dialysis and freeze drying . The Polysaccharide contents in the sample of Hericium polysaccharides was 75.2%(±0.64%),while in the sample of Lentinus edodes polysaccharides was 80.0%(±0.63%).Not only was polysaccharide in the samples pure which was detected by high performance liquid chromatography analysis , but also the polysaccharide of beta-pyrano polysaccharides which was detected by infrared spectrum analysis . Beta-glucan contents in the sample of Hericium polysaccharides was 64.94%(±1.42%),while in the sample of Lentinus edodes polysaccharides was 70.62%(±0.24%). Hericium erinaceus polysaccharide was composed by four kinds of Monosaccharide named galactose、glucose 、xylose and mannose , while five sorts of Monosaccharide named arabinose、galactose 、glucose、 xylose and mannose were detected in Lentinus edodes polysaccharide by the method of Ion chromatography . Protein content detected by the method of BCATM kit in Hericium erinaceus polysaccharide was 4.84%(±0.12%), while 8.68%(±0.20%)in Lentinus edodes polysaccharide . It was suggested that the two polysaccharides had antitumor effect after observing by putting Hericium erinaceus polysaccharide and Lentinus edodes polysaccharide respectively with RAW 264.7 macrophage to co-cultured for a period of time.
