摘要目的:研究腺梗豨莶水提取物对3T3-L1脂肪细胞的增殖抑制和胰岛素抵抗的作用。
方法:用MTT法检测腺梗豨莶水提取物对3T3-L1脂肪细胞的毒性;用油红O染色法检该提取物对成熟的3T3-L1脂肪细胞的增殖抑制作用;建立胰岛素抵抗模型,用葡萄糖氧化酶-过氧化物酶法测定培养液中葡萄糖的水平,检测该提取物对胰岛素抵抗的作用。89412
结果:MTT检测结果表明腺梗豨莶水提取物作用的最佳时间是48 h,且在100 μg/ml以下的浓度无细胞毒性。油红O染色结果表明该提取物在诱导成熟的3T3-L1脂肪细胞48 h后,50 μg/ml浓度的水提取物与对照组相比具有显著性差异;该提取物经过反向OD柱分离所的流分(XXC-A1,A6)与对照组相比也具有显著性差异。但是,葡萄糖水平检测结果表明水提取物与对照组相比无显著性差异。
结论:50μg/ml的腺梗豨莶草水提取物及其所经反向ODS分离的流分(XXC-A1,A6)对成熟的3T3-L1脂肪细胞增殖有显著抑制作用。但是,腺梗豨莶水提取物在胰岛素抵抗模型中无降低葡萄糖水平的作用。

Abstract
Objective: To study the inhibitory effect of water extract of Siegesbeckia pubescens Makino on adipocyte proliferation and insulin resistance。
Methods: The toxicity of water extract of S。 pubescens Makino in 3T3-L1 cells was detected by MTT method。 The Inhibitory effect on the proliferation of mature 3T3-L1 adipocytes was evaluated by Oil red O staining。 The model of insulin resistance was established。 The glucose level in the culture medium was determined by glucose oxidase - peroxidase method, and the effect of the water extract of S。 pubescens Makino on insulin resistance was tested。
Results: The optimal time for the action on water extract of the S。 pubescens Makino was 48 h and there is no cytotoxicity at the concentration of 100 μg/ml or less。 There is a significant difference at the 50μg/ml of the S。 pubescens Makino water extract compared with the control group after inducing mature 3T3-L1 adipocytes 48 h。 What’s more, the two fractions(XXC-A1,A6) isolated from water extract of S。 pubescens have significant differences compared with the control group。 Unfortunately, there is no significant difference of the S。 pubescens Makino water extract in the glucose level。
Conclusion: The water extract of S。 pubescens Makino at 50 μg/ml and the two fractions (XXC-A1,A6)isolated from it by ODS have significant inhibitory effects on adipocyte proliferation in 3T3-L1 cells。 The aqueous extract of S。 pubescens Makino do not decrease the glucose level in insulin resistance model。
