    摘要锦灯笼,又名挂金灯、红姑娘,是茄科酸浆属的草本植物。锦灯笼宿萼苦,果实甘,鲜果可以作为水果生食,全草及其果实都可入药,具有利咽、清热、解毒、化痰、利尿的作用。锦灯笼在我国许多地区均有分布,药源丰富。现代药理学研究已证明其具有抗肿瘤活性。50438

    本研究以锦灯笼带果实的宿萼作为研究对象,采用生物活性追踪法,即采用噻唑蓝(MTT)比色法有针对性地从锦灯笼中分离鉴定抗肿瘤活性部位及其活性成分。研究结果表明,锦灯笼二氯甲烷和石油醚萃取物对人肺癌细胞 A549、人前列腺癌细胞 PC-3、人乳腺癌细胞 MCF-7 和人肝癌细胞 HepG2 均有不同程度的增殖抑制作用,其中二氯甲烷部位对这 4 种肿瘤细胞的毒性更强,初步确定二氯甲烷萃取层为锦灯笼的抗肿瘤活性部位。

     二氯甲烷萃取物经第一次硅胶柱色谱分离,得到12个流分,其中流分4、5对肿瘤细胞的增殖抑制作用最明显。对流分5进行第二次硅胶柱色谱分离,得到具明显抗肿瘤活性的流分E。对流分4进行两次硅胶柱色谱分离得到流分P,采用制备反相液相色谱从中制备得到化合物Q,经鉴定为睡茄内酯类化合物中的withaminimin。

    Abstract Physalis alkekengi L. var. franchetii (Mast.) Makino, also known as Guajindeng, Hongguniang, is a herb belonging to Solanaceae family and Physalis species. It has bitter calyxes and sweet fruit. The raw fruit can be directly eaten as a fruit. The whole plant and its fruit can be used as medicine. This herb can relieve sore throat and has effect of detoxification, phlegm resolving and diuresis. The herb is widely distributed in China. Modern pharmacology researches have proven its anti-tumor activity.

    In this study, bioactivity-tracing method (MTT colorimetric method) was used to screen the active fractions and ingredients from its fruit and calyxes. The results showed that both dichloromethane and petroleum ether extracts had certain degree of growth inhibition on human lung cancer cell line A549, human breast cancer cells MCF-7, human prostate cancer cells PC-3 and human hepatoma cells HepG2. Furthermore, cytotoxic activity of the dichloromethane extract was stronger than that of the petroleum ether extract. In conclusion, the dichloromethane extract was demonstrated to be the major bioactive fraction of Physalis alkekengi var. franchetii.

    With the first-run silica gel column separation, 12 fractions from the dichloromethane extract were collected and the fraction 5 had the highest anti-tumor activity in vitro, followed by the fraction 4. The second-run silica gel chromatography was performed with the fraction 5, and the fraction E was found to have the most significant proliferation inhibition towards these tumor cells. After twice silica gel chromatography was performed with the fraction 4, we obtained a fractions P. A compound Q was separated and purified from the fractions P using preparative high performance liquid chromatograph. It was identified as a withanolide type compound, named as withaminimin.
