摘  要植物内生真菌是指与植物组织共生存,不影响植物组织功能效果的真菌,研究表明,植物内生真菌具有抗肿瘤的生物活性。

本课题所采取的样本来自于日常生活中的普通植物花生,从花生标本中分离出内生真菌,对其进行纯化,经鉴定,该内生真菌是拟茎点霉。对拟茎点霉内生真菌进行发酵培养后,检测其代谢产物的抗肿瘤活性。然后对拟茎点霉发酵液进行内生真菌抗肿瘤活性物质的粗提取。通过多种发酵液预处理方法的比较,如加热法、调节pH值法、无水乙醇直接沉淀法和(NH4)2SO4分级盐析法, 确定加热法是拟茎点霉发酵液抗肿瘤活性物质的最佳粗提取方法。为了进一步分离纯化得到抗肿瘤活性物质,需要对发酵液理化性质进行初步研究。通过溶媒萃取试验、层析试验、纸电泳试验和稳定性试验等方法,得出拟茎点霉发酵液中抗肿瘤活性物质是一种弱极性强碱性的水溶性物质,在光照、高温及酸碱条件下均稳定,有利于该抗肿瘤活性物质药物的保存。69838

Preliminary isolation of antitumor active substance from Phomopsis sp.

Abstract

The plant endophytic fungi exist in the plant tissues, which can not affect the function of the plant tissues. The study shows that the endophytic fungi have the biological activity against the tumor.

The sample used in this topic came from the plant peanuts. The endophytic fungi was isolated from the peanut samples and purified. It was identified that the endophytic fungi was Phomopsis sp.. After fermentation cultivation on Phomopsis sp., we tested its antitumor activity. Then the crude extract of the fermented liquor was extracted. Comparing a variety of fermented liquor pretreatment methods, such as heating method, adjusting pH value method,  anhydrous ethanol method and (NH4)2SO4 fractional salting out method, we found that the heating method was the best crude extraction method. We need to study the physical and chemical properties of Phomopsis sp. for getting the antitumor active substance. By solvent extraction test, chromatography test, paper electrophoresis test and stability test, it was concluded that the antitumor active substance was a kind of weak polarity strong alkaline water-soluble substance. It was stable under light, high temperature and acid-base for the preservation of the antitumor active substance.
