摘要为了研究分离植物中的抗肿瘤活性物质,本课题从红薯标本中分离出多种植物内生菌,对其进行纯化,并进行发酵培养后,检测其抗肿瘤活性。
红薯,又名番薯,红薯叶,顾名思义就是红薯生长过程中的茎上的叶子,经研究发现红薯叶中含有极高的维生素、矿物质元素和蛋白质,亚洲蔬菜研究中心已经把红薯叶列为高营养蔬菜。据研究发现,红薯叶中含有黄酮、绿原酸、多糖 、多肽等活性物质,这些物质具有抗肿瘤的作用。我们从红薯叶中分离出的一株具有抗肿瘤活性的内生真菌,经鉴定为软毛毛壳菌。本文通过对软毛毛壳菌进行加热法、调pH值法、(NH4)2SO4 分级盐析法等方法对菌株的活性物质进行粗提取,经实验结果检测数据显示,(NH4)2SO4 分级盐析法能较好地用于初步去除杂质。接着通过溶媒萃取,纸电泳,稳定性试验等方法对软毛毛壳菌进行理化性质分析,为进一步的分离纯化作准备。72221

The Isolation and Extraction of antitumor active substances of Chaetomium mollipilium
Abstract
To study the isolation of antitumor active substances from plants, the topic isolated many kinds of endophytic fungus from sweet potato samples。 After the purification of these endophytic fungus, fermenting it and detecting its antitumor activity。
Sweet potato is a kind of food we familiar。 Sweet potato leaves are the leaves on the stem during the growth process, the study found that sweet potato leaf contains extremely high vitamins, minerals and proteins, Asian Vegetable Research Center has taken the sweet potato leaf as high nutrient vegetable。 Study found that the sweet potato leaves contain flavonoids, chlorogenic acid, polysaccharide, polypeptide and other active substances, which have anti-tumor effect。 After identification, we isolate a Chaetomium mollipilium with antitumor activity from sweet potato leaf。 We use a variety of methods to extract the active substance of the strain, such as heating method, acid base method, (NH4)2SO4 salting out and so on。 According to the data, (NH4)2SO4 salting out method is better than other methods。 Then, we study its physicochemical properties by solvent extraction, paper electrophoresis, stability test and so on, for further isolation and extraction。
