摘要间充质干细胞(bone marrow stromal cells, BMSCs)是一类有多向分化潜能和极高医学价值的细胞。但随着细胞的衰老,其分化能力受到影响,临床治疗效果不理想,因此,急需找到抑制间充质干细胞衰老的方法。研究表明 ,紫甘薯花青素具有明显的抗氧化活性,但其在间充质干细胞老化中的作用尚不明确。本实验利用H2O2预处理诱导BMSCs老化,经不同浓度的紫甘薯花青素处理后,发现紫甘薯花青素可以抑制氧化压力引起的BMSCs老化;鉴于细胞自噬与老化之间的关联,我们又检测了自噬相关指标LC3的聚集情况。结果显示,H2O2可以降低LC3的聚集分布,表明自噬被抑制;而紫甘薯花青素可以增强LC3的点状聚集,即诱导细胞自噬。这一结果证实,紫甘薯花青素可以通过促进细胞的自噬来抑制间充质干细胞的衰老。50706

Inhibitory Impact of Purple Sweet Potato Color on Bone Marrow Stromal Cells Senescence
Abstract
Bone marrow stromal cells (BMSCs) are kinds of cells with pluripotent differentiation and high medical value. However, their differentiation ability is impaired during cell senescence, which limits the application in clinic. Therefore, it is urgent to find methods to inhibit BMSCs senescence. Papers have demonstrated the antioxidation of purple sweet potato color (PSPC), but little is known about its function in BMSCs senescence. To answer this question, BMSCs were pre-incubated with H2O2 and then administered with PSPC. Senescence-associated β-gal staining showed that PSPC could delay BMSCs senescence induced by oxidative stress and this effect was dose-dependent. Based on the relation between autophagy and senescence, we further detected the changes of LC3 puncta. The results showed that H2O2 decreased the numbers of puncta, indicating autophagy was inhibited by H2O2; On the contrary, PSPC promoted the patched distribution of LC3, suggesting PSPC could induce cell autophagy. In conclusion, our data indicated that PSPC could inhibit the senescence of BMSCs by promoting cell autophagy.
