摘  要癌症是近年来发病率和死亡率持续升高的疾病,尤其是乳腺癌严重危害女性身心健康,有效药物却极其短缺,因此,新的抗癌药物的开发迫在眉睫。放线菌次生代谢产物丰富,从中筛选新的抗癌药物备受人们关注。本课题组前期从来自新疆格尔木地区的土壤中分离得到一株能产生抗肿瘤活性的放线菌,命名为G3。本研究在前期工作基础之上,通过划线法纯化G3菌株,使用大豆玉米培养基进行发酵培养,检测其代谢产物,发现具有良好的抗肿瘤活性;使用硫酸铵盐析、透析以及萃取、浸提等方法对发酵产物进行初步分离,以对抑制乳腺癌细胞活性为指标,发现分离得到的大分子物质活性显著高于小分子物质。同时,对G3放线菌的发酵培养基进行筛选,分别使用大豆玉米培养基、高氏一号合成培养基、PD培养基,以及ISPII培养基,通过对G3放线菌菌丝体不同形态的观察、发酵产物蛋白浓度检查、抗肿瘤活性检测,分析确定高氏一号合成培养基为G3放线菌发酵的最佳培养基。本研究为G3放线菌抑制肿瘤细胞活性成分的分离纯化奠定基础。69845

Antitumor Activity of Actinomycete G3 Metabolites 

Abstract Cancer has become a high incidence and mortality disease in recent years, which continues to rise at a high speed. Especially, breast cancer is a serious health hazard in women. But extremely effective drugs are limited. Therefore, the development of new anti-cancer drugs is imminent. Actinomycetes can produce rich secondary metabolites, which has made screening of new anti-cancer drugs much attention. The actinomycete called G3 used in this experiment was isolated from the soil of Golmud region in Xinjiang. On the basic of previous research, I purified actinomycete G3 saved in the key laboratory by scribing method firstly, then used the corn and soybean medium to measure the antitumor activity and its metabolites was found to have good anti-tumor activity. The inhibition activity of breast cancer cell was assessed as an index in this experiment and the methods such as ammonium sulfate precipitation, dialysis and extraction were used for fermentation products’ preliminary separation. I found that the activity of isolated macromolecules was significantly higher than small molecules. At the same time, the fermentation media for actinomycetes G3 was responsible for screening, respectively including corn and soybean medium, Goates synthetic I medium, PD and ISPII medium. Through the observation of different media in different forms mycelium, detection of protein concentration in fermentation product, and measure in anti-tumor activity, I made sure that Goates synthetic I medium was optimal. The researches laid the foundation for the isolation and purification of anti-tumor ingredient from actinomycetes G3 metabolites.
