摘要:乙酰胆碱酯酶是一种多分子型繁杂的蛋白质,大多数存在于胆碱能神经末梢突触间隙。本课题对乙酰胆碱酯酶进行固定化研究,为乙酰胆碱酯酶抑制剂筛选新方法奠定基础。通过实验将酶与载体(SBA-15)固定化,探究了不同添加剂,交联剂对固定化酶热稳定性的影响,得到一种热稳定性较高的固定化酶固定化方法。实验结果显示最佳固定化条件为:戊二醛20 mM,固定化过程中固定含有0.3%蔗糖、0.3%BSA和0.4%明胶混合添加剂的固定化酶具有最佳的酶稳定性。同时,该固定化酶在4 oC和25 oC下热浴2小时,剩余酶活可达90%以上,因此,该固定化酶有望能够在室温下长期储存。71237

Acetylcholinesterase’s molecular simulation and immobilization on SBA-15
Abstract: Acetylcholinesterase is a multi-molecular protein, which is very complicated. It mainly appears in the cholinergic nerve endings of synaptic space. Our group prepared to immobilize the acetylcholinesterase as the base for acetylcholinesterase inhibitors. By combining the enzyme with the vector (SBA-15), we intended to find out the effects of different additives and crosslinking agents on the properties of immobilized enzymes. Finally, we obtained a method with high thermal stability for immobilizing the enzyme. In conclusion, the optimal immobilization condition is achieved: Glutaraldehyde 20mmol, containing 0.3% sucrose, 0.3% BSA and 0.4% gelatin mixed additives in the immobilization with low temperature remained the best enzyme activity. At the same time, the remaining enzyme activity was over 90% which was heated at 4oC and 25 oC for 2 hours, so that the immobilized enzyme is expected to be stored at room temperature for a long period.
