摘要目的:利用纸层析法初步分离一株内生真菌代谢产物粗提物中的抑菌活性成分。方法:对内生真菌发酵液进行粗提取,用微量肉汤稀释法验证粗提物的抑菌活性。采用捷克八溶剂系统对粗提物进行纸层析,初步分离其抑菌活性成分。结果:用微量肉汤稀释法验证了内生真菌粗提物的抑菌活性;在纸层析实验中,从8个展层系统中挑选出了5个分离效果较好的展层系统,分别是:4号、5号、6号、7号、8号展层系统。将这5个展层系统进一步进行不同点样量的实验,发现当点样量为10ul,在7号系统(苯:甲醇=4:1)中分离效果最好,条带上7-10cm处含有抑菌活性成分,推测抑菌活性成分的Rf值为:0。46-0。66。本研究为进一步分离纯化内生真菌代谢产物的抑菌活性成分奠定基础。72225

Study on the Primary Separation of the Antibacterial Components from A Strain of Endophytic Fungus by Paper Chromatography
Abstract Objective: To preliminarily separate the antibacterial components from the crude extract of metabolites of an endophytic fungus by paper chromatography。 Methods: The endophytic fungus was cultured in the Potato dextrose liquid medium and the crude extract of the fermentation broth was obtained。 The antibacterial activity of the extract was determined by the micro-broth dilution method。 Czech eight solvent system was chose as developing agents on paper chromatography for the primary separation。 Results: The antimicrobial activity of the extract was verified by the micro-broth dilution method。 In the paper chromatography experiment, five developing systems with better separation effect were picked out from all the eight systems, which were NO。4, 5, 6, 7 and 8 systems。 The five systems are further tested the separation effect with different extract volumes sampled。 It was found that the best one was 7 system (benzene: methanol =4:1) when the sample quantity is 10ul。 The paper segment which was 7-10cm from the sampling place contained the antibacterial active components, suggesting that the Rf value of the antibacterial activity component is range from 0。46 to 0。66。This study laid a foundation for further isolation and purification of the antimicrobial activity of endophytic fungal metabolites。
