摘 要目的:本研究以 AD 小鼠模型(APP/PS1)为实验对象,研究亚精胺(Spermidine,Spd) 对 AD 小鼠脑组织病理及淀粉样斑块形成的影响。方法:对 AD 小鼠进行亚精胺干预,每周 两次,持续 26 周;AD 小鼠取脑制作石蜡切片;用 anti-Aβ 抗体进行免疫荧光染色,检测 AD 小鼠 Aβ 淀粉样斑块,通过 Image J 软件测量斑块的直径,Excel 表格统计斑块数量和计 算斑块面积;运用免疫组织化学技术和刚果红染色,研究亚精胺对淀粉样斑块形成的影响; 亚精胺对 AD 模型小鼠细胞自噬的作用;亚精胺对 AD 模型小鼠自噬关键蛋白 Beclin 1 的作 用。结果:AD 小鼠刚果红染色显示海马和皮层区有粉红色斑块。皮层区斑块面积>2000µm2、1500-2000 µm2、1000-1500 µm2 的 AD NS 组和 AD Spd 组斑块数量有显著性差异;海 马区斑块面积>2000 µm2 的 AD NS 组和 AD Spd 组斑块数量有显著性差异;亚精胺增加 AD 小鼠中 P62 底物的清除,减少 LC3 深染的细胞数,下调斑块周围 Beclin 1 的入核。结论: 亚精胺能减少 AD 小鼠海马区面积大于 2000 µm2 的斑块数,减少皮层区面积大于 2000µm2、1500-2000 µm2、1000-1500 µm2 的斑块数。亚精胺改善 AD 小鼠脑内自噬降解功能障 碍,缓解斑块周围 Beclin 1 的核内移。88814

Abstract Objective: The research focuses on AD mice model (APP/PS1) for the experiment subject。 In order to study the effects of spermidine on brain pathology and amyloid plaque formation in AD model mice。 Methods: Giving an intervention of spermidine on AD mice twice a week last 26 weeks; Get brain from AD mice to make paraffin section; Immunofluorescence staining with anti-Aβ antibody to detect amyloid plaques in AD mice。 Measure the diameter of the plaques through Image J software, quantity of plaques and patch area are counted by Excel spreadsheet; Immunohistochemical technique and Congo red stain are used to research the formation of amyloid plaques。 Investigate the effects of spermidine on amyloid plaque formation, the role of spermidine in autophagy on AD mice and the impact of
spermidine on Beclin 1。 Result: AD mice which stained with Congo red possess pink patches in hippocampus and cortex, the plaque area which greater than 2000 µm2、 1500-2000 µm2 、1000-1500 µm2 in cortex between AD NS and AD Spd has significant difference; Plaque area which greater than 2000 µm2 in hippocampus
between AD NS and AD Spd has significant difference。 Spermidine increases the clearance of the P62 substrates, reduces the number of LC3 hyperchromatic cells, decrease Beclin 1 into the nucleus。 Conclusion: Spermidine can reduce plaque number which area greater than 2000 µm2 in hippocampus on AD mice, and decrease
plaque number which area greater than 2000 µm2、1500-2000 µm2、1000-1500 µm2
in cortex on AD mice。 Spermidine improves autophagy degradation dysfunction in the brain of AD mice, alleviates Beclin 1 into the nucleus around the plaque。
