Cloning of LeEIN2 Gene and Vector Construction of Gene Silencing from Tomato
Abstract: EIN2, a key factor of the ethylene signal transduction system, was the first positive regulatory factor, which located downstream of receptor/CTR1 complex. This research focuses on constructing virus induced gene silencing (VIGS) vectors of EIN2. Firstly, total RNA was extracted from tomato, and then EIN2 gene was cloned by reverse transcription RT-PCR. Secondly, EIN2 was constructed to pMD18-T vector, afterwards the recombinant pMD18-T-EIN2 vectors we constructed were successfully identified by PCR and sequencing. Finally, EIN2 was constructed to tobacco rattle virus (TRV) vector (pYY13), after that the positive recombinant pYY13-EIN2 vectors were successful by sequencing and PCR identification. The research results laid the foundation for the function research of EIN2.
