摘要:应用DNS法测定木霉T-soybean的几丁质酶活性,结果表明,添加几丁质诱导的比不添加几丁质诱导的木霉T-soybean几丁质酶活性要高,而且在一定范围内酶活性随几丁质浓度的增加而增高,当几丁质浓度超过0.9g/mL后酶活性随几丁质浓度的增加而降低;在添加几丁质诱导的情况下第三天的酶活性是最高的;在不同pH值,等量几丁质诱导的情况下,pH=6.0时几丁质酶活性是最高的;应用平板对峙法来检测几丁质酶对尖孢镰刀菌和核盘菌的抑制作用,实验结果表明,几丁质酶对尖孢镰刀菌和核盘菌的生长均有一定程度的抑制。28530 毕业论文关键词:几丁质酶;木霉T-soybean;酶活性
Analysis of Trichoderma soybean Chitinase Function
Abstract:Chitinase activity of Trichoderma T-soybean was determined by DNS method. Enzyme activity induced by chitin was much higher compared to that of without chitin induction. Chitinase enzyme activity increased with the increasing of chitin concentration in a certain range. However, enzyme activity decreased when chitin concentration was more than 0.9g/mL. Meanwhile, chitinase activity was the highest after culture three days at the chitin concentration of 0.9g/mL, as well as pH=6.0. The inhibitory effects of chitinase on Fusarium oxysporum and Sclerotinia sclerotiorum were determined. It was showed that chitinase had a certain degree of inhibition on F. oxysporum and S. sclerotiorum.
