摘要:EIL(Ethylene-insensitive 3-like)是乙烯信号通路中重要的转录因子。本实验发EIL共抑制转基因番茄(EIL-CS line)出现单性结实现象,经测定发现其单性结实果实的内源生长素浓度明显高于EIL过表达和野生型对照植株,说明抑制EIL的转录能够提高开花后子房中生长素含量,从而引起单性结实。为了进一步探究乙烯与生长素在果实形成过程中的互作关系,本实验通过研究EIL-OX、pEIL-GUS和DR5-GUS等转基因番茄,分析了EIL蛋白和生长素在花蕾中的表达和合成规律,结果显示,开花前期,EIL超表达(EIL-OX)番茄中生长素生物合成高于野生型对照,开花后生长素的合成无明显差异。生长素在乙烯信号关键因子EIL的影响下,在开花前后表现出不同的合成规律,说明开花前后EIL与生长素的互作机制存在不同。29786

Study on the Interaction of Ethylene and Auxin in Tomato Fruit Development
Abstract:EIL (Ethylene-insensitive3-like) is an important transcription factor in the ethylene signaling pathway. In this study, Parthenocarpic fruit set was observed in EIL co-suppressed transgenic tomato (EIL-CS line). The concentration of endogenous auxin in the fruit of EIL- CS was significantly higher than that of EIL overexpression and wild type control plants, which indicated that inhibiting the transcription of EIL could increase the content of auxin in the ovary after flowering. In order to further understand the interaction between ethylene and auxin in the process of fruit formation, the expression pattern of EIL and synthesis of auxin in buds were studied via EIL-OX, pEIL-GUS and DR5-GUS transgenic tomatoes. The results showed that the auxin biosynthesis of EIL-OX was higher than that of wild type control, but there was no significant difference in the synthesis of auxin after flowering. Under the influence of EIL, the auxin showed different synthetic characteristics before and after flowering, indicating that the mechanism of interaction between EIL and auxin was different before and after flowering.
