摘要:橙花醇(Nerol),无色油状液体,香柔和优美,又相对偏清,是珍贵的香精和医药中间体,在香精、医药、食品、日化等行业被普遍使用。白色念珠菌,又名白色假丝酵母菌,酵母科念珠菌属,是常见的条件致病性真菌。本实验主要参照美国国家临床实验标准化委员会(NCCLS)制定的真菌敏感性测定方案(M38-A2),采用微量稀释法测定橙花醇对白色念珠菌的MIC为0.05 µL/ mL,通过紫外分光光度计测定麦角固醇含量的变化,并应用流式细胞仪检测橙花醇对白色念珠菌细胞膜的损伤和橙花醇对其细胞周期和细胞凋亡的影响,结果表明,橙花醇能通过影响白色念珠菌的对数生长期细胞的生长,抑制其生长,在白色念珠菌的细胞凋亡上,橙花醇通过使其膜内磷脂酰丝氨酸外翻,导致细胞凋亡。34360 毕业论文关键字:橙花醇;白色念珠菌;细胞周期;细胞凋亡
Effects of Nerol on the Cell Cycle and Cell Apoptosis of Candida albicans
Abstract:Nerol, a colorless oily liquid, smell beautiful and soft, and relatively clear, is a valuable spice and pharmaceutical intermediates, in essence, medicine, food, cosmetic and other industries has extensive application value. C. albicans, also known as white Candida yeast, is a common condition pathogenic fungi. The microdilution was applied to detect MIC of C. albicans which treated by nerol according to M38-A2 method of NCCLS, the MIC is 0.05 µL/mL. The change of ergosterol content was determined by ultraviolet spectrophotometer. The cell cycle and apoptosis of C. albicans treated by nerol was detected by flow cytometry. the results show that the nerol can inhibit the growth of C. albicans by effect it’s logarithmic growth phase cells, and on the cell apoptosis, nerol lead to apoptosis by make the phosphatidylserine outside the membrane.

引言
现今,由于免疫抑制剂、广谱抗生素和细胞毒药物的大量应用;器官移植、静脉营养和导管介入等治疗手段的普及;糖尿病、恶性肿瘤、血液病、艾滋病、老年病等临床严重疾病的发生率的不断上升[1],使得真菌感染,尤其是条件真菌感染的发病率及死亡率逐年上升[2]。
白色念珠菌(C. albicans),又名白色假丝酵母菌,酵母科念珠菌属,是常见的条件致病性真菌[3],是人体皮肤、粘膜的正常寄生菌,可引起人黏膜、皮肤、消化道、呼吸及神经系统等感染[4],人体免疫力低下时会引起浅表性的或深部的感染,严重者会引起系统性感染,将危及生命[5]。
