摘要:本课题的研究目的旨在探究OsNAC300是否参与水稻响应镉(Cadmium, Cd)胁迫应答。以20天苗龄的的水稻幼苗为材料,我们用0、0.5、5和50 μM四种浓度镉处理3天,并测定水稻的生长指标和镉含量。实验结果表明:与野生型相比,过表达OsNAC300的水稻幼苗的生长速度更快,生物量更大,但耐镉性及镉含量没有显著性差异。另外,又以OsNAC300p:GUS转基因水稻胚根为材料,我们使用加镉和不加镉条件处理24小时,然后进行GUS染色和体视显微镜拍照。GUS染色结果显示镉胁迫会显著诱导根中OsNAC300基因的表达。总之,我们发现:一方面,OsNAC300可能仅在转录水平响应镉胁迫应答,但不影响水稻对镉的吸收和耐性;另一方面,OsNAC300可能参与对水稻幼苗生长进程的调控。38752 毕业论文关键词: OsNAC300;过表达;镉胁迫
Research on Function of Cadmium Stress Responsive Transcription Factor OsNAC300 in Rice
Abstract: The purpose of this study was to investigate whether OsNAC300 is involved in the response of rice to cadmium (Cadmium, Cd) stress. 20 days-old Rice seedlings were treated with solutions containing 0, 0.5, 5 and 50 μM cadmium for 3 days, and the growth index and cadmium content were determined. The rice seedlings which overexpressing OsNAC300 showed higher growth rate and larger biomass compared with wild type, but there was no significant difference in cadmium tolerance ability and cadmium content. In addition, using OsNAC300p:GUS transgenic rice as material, we treated rice seedlings for 24 hours using +Cd and -Cd conditions, and carried out GUS staining and microscopy observation. The results of GUS staining showed that cadmium stress significantly induced the expression of OsNAC300 in roots. In conclusion, we found that: OsNAC300 may only respond to cadmium stress at the transcriptional level, but does not affect the uptake and tolerance of rice to cadmium; we suggest OsNAC300 may be involved in the regulation of the growth process of rice seedlings.
