摘要  随着心血管疾病成为影响人类健康的最大阻碍,对心血管疾病的预防越来越迫在眉睫。 由于内皮细胞的老化与心血管疾病的发病密切相关,我们拟通过探究紫甘薯花青素(purple sweet potato color,PSPC)抑制内皮细胞老化的作用来找寻突破口。在本实验中,我们将处于对数生长期的人脐静脉血管内皮细胞,分为三组,分别为正常组,高糖组和 PSPC 组,处理 24h 和 48h;对内皮细胞进行衰老相关的 β-Gal 染色;酶联免疫吸附反应法(ELISA)检测细胞分泌炎性因子的能力;一氧化氮(NO)试剂盒检测NO浓度;流式细胞仪检测细胞周期。β-Gal 染色结果显示 PSPC 抑制高糖诱导的内皮细胞老化,ELISA 检测结果显示 PSPC 抑制内皮细胞炎性因子的分泌,NO 试剂盒检测结果表明 PSPC 通过提高 NO 活性进而恢复内皮细胞功能,细胞周期实验显示 PSPC 可以逆转高糖引起的 G1 期阻滞。通过以上实验,我们得到结论:PSPC 对高糖诱导的内皮细胞衰老有显著的抑制作用,其机制可能与 PSPC 影响细胞周期,抑制炎性因子分泌和恢复细胞功能有关。 该论文有图6 幅,表1个,参考文献20 篇。  50767

Since endothelial senescence is significantly related to cardiovascular disease, we try to find a breakthrough by exploring the inhibitory effect of purple sweet potato color(PSPC)on senescence of endothelial cells. In this study, human umbilical vein endothelial cells in logarithmic phase were pided into three groups for 24h and 48h :normal group, high glucose group and PSPC group ;senescence of endothelial cells was identified by β-Gal staining ;secretion of inflammatory cytokines was detected by enzyme-linked immunosorbent assay(ELISA); NO expression was tested by NO kit; cell cycle was analyzed by flow cytometry . The result of β-Gal staining showed PSPC suppressed high glucose-induced endothelial cell aging .Meanwhile, PSPC inhibit the secretion of inflammatory cytokines in endothelial cells as showed by ELISA test, but endothelial function was restored by PSPC via increasing NO concentration. Therefore, we got the conclusions : PSPC inhibited high glucose-induced senescence of endothelial cells, which may be related with cell cycle, inhibition of inflammatory cytokine secretion and restore cell function.
