摘要:灵芝(Ganoderma Lucidum Karst)又称赤芝、红芝、木灵芝、菌灵芝、灵芝草。具有止咳平喘,补气安神的功效,用于虚劳咳喘,眩晕不眠,心悸气短。灵芝所含化学成分复杂,其所含的化学成分因所用菌种、培养方法、提取工艺的不同而变化。研究表明,灵芝中除了含有三萜类化合物和多糖类化合物外,还含有甾醇类、核苷类、生物碱类、呋喃衍生物、氨基酸多肽类、无机元素和脂肪酸等化学成分,但灵芝三萜和灵芝多糖是灵芝中的主要的活性物质。研究灵芝的化学成分的目的不仅在于了解和比较灵芝属不同品种及同一品种的不同发育阶段所含的化学成分的差异,还可以通过提取的方法确定其有效活性成分和含量。20160
本研究通过前阶段对沪农1号50KG灵芝子实体进行了中试提取,取部分提取物用50%,60%,70%,80%,90%,100%乙醇溶液在D-101、HZ-801、HZ-818三种大孔树脂做分段洗脱实验,发现最适合富集灵芝三萜化合物的大孔树脂为D-101型大孔树脂,同时将各分段洗脱下来的提取物进行了高效液相色谱分析和抗肿瘤活性测定。分析结果表明三萜化合物主要集中在90%和100%醇提灵芝液中,且这部分的洗脱物具有很强的细胞毒性。随后进行了大孔树脂的中试洗脱,最终获得了富含三萜化合物的组分。将这一组分进行了硅胶柱层析,获得了麦角甾醇、灵芝酮三醇两个化合物。

The active ingredients in Ganoderma lucidum extraction and enrichment technology research
Abstract: Ganoderma Lucidum Karst was also known as Ganoderma lucidum, ganoderma,Jun Lingzhi, and lucid ganoderma, which has the efficacy of relieving cough and asthma, conserving vitality, and soothing the nerves. It treats the diseases like consumptive cough, dizzy insomnia, palpitations, and shortness of breath. The chemical composition of G. lucidum is complex. Its chemical composition is different by the species, culture method, and extraction process. The research shows that the G. lucidum has triterpenoids and polysaccharide compounds besides sterols, nucleoside, alkaloids, furan derivatives, amino acid polypeptide, inorganic elements and fatty acids, etc., but triterpenoids and polysaccharides were the main active substance in G. lucidum. The purpose of the research is not only to understand the chemical composition difference between the different and same species of the G.lucidum in different development phase, but also to make sure the active ingredient and content of the G. lucidum by extraction method.
The study was through the Pilot Extraction of the Hunong number 1 G.lucidum in fifty kilogram prior stage.  Then take parts of the extract with 50%, 60%, 70%, 80%, 90%, and 100% ethanol solution into D-101, HZ-801, and HZ-818 macroporous resin respectively. The discovery of the elution experiments was that the most suitable macroporous resin to enrich the G.Lucidum triterpenoid was D-101 macroporous resin. And then use each segment eluting extract through High Performance Liquid Chromatography analysis and antitumor activity test. The result of analysis showed that the triterpenoid mainly in 90% and 100% alcohol extract of G.lucidum liquid, and the eluate of this part had strong cytotoxic activity. The following experiment was to elute macroporous resin, which finally obtained the component filled with triterpenoid. And in the end take the component into silica gel column chromatography, which achieved two compounds ergosterol and ganodermanontriol.
